| Objective To evaluate the value of serum mi R-92a-3p in the diagnosis and prognosis of colorectal cancer and to compare the sensitivity of serum mi R-92a-3p with traditional test of CRC screening.Methods 1. Serum total RNA was extracted from 30 healthy volunteers, 25 adenomatous polyps patients and 40 preoperative and postoperative CRC patients. Levels of mi R-92a-3p were determined by FQ-PCR. 2. The differences of mi R-92a-3p levels among four groups and the relationship between serum mi R-92a-3p levels and clinical-pathological characteristics of CRC patients were analyzed by Kruskal-Wills H test. 3. The ROC curve was drawn in order to evaluate the diagnostic efficiency of serum mi R-92a-3p in patients with colorectal cancer. 4. The fecal occult blood and serum carcinoembryonic antigen levels of 40 CRC patients(including preoperative and postoperative CRC patients) were detected in order to compare the sensitivity of serum mi R-92a-3p and traditional test of CRC screening.Results 1. The relative expression levels of serum mi R-92a-3p in preoperative CRC patients were significantly higher than those postoperative CRC patients, patients with adenomatous polyps, and healthy volunteers(Z=4.75, 4.28 and 8.00, respectively, all P<0.01).The relative expression levels of serum mi R-92a-3p in healthy volunteers were significantly lower than those in the CRC patients after surgical resection(Z=4.08, P<0.01) and the patients with adenomatous polyps(Z=2.70, P=0.01),while no significant difference between the later two(Z=0.99, P=0.32). 2. The area under the ROC curve(AUCROC) of serum CEA for the diagnosis of CRC was 0.608(95%CI: 0.491-0.726).The area under the ROC curve(AUCROC) of serum mi R-92a-3p for the diagnosis of CRC was 0.899(95%CI: 0.840-0.959).When the cut off value was set as 2.75, both of sensitivity and specificity were 80%. 3. The expression levels of mi R-92a-3p were significantly associated with the depth of tumor invasion(Z=2.14, P=0.03), TNM stage(Z=2.93, P<0.01) and lymph node metastasis(Z=2.86, P=0.04). 4. CEA levels of preoperative and postoperative CRC patients are respectively 2.88(1.30, 17.60)and 3.55(1.32, 16.87)and no significant difference was observed(Z=0.192, P=0.847). The total positive rate of FOBT of CRC patients was 69.6%。Conclusion Sensitivity of serum mi R-92a-3p is higher than tranditional screen methods. It may be use as the diagnostic and prognostic biomarkers for CRC patients. Serum mi R-92a-3p may promote the invasion and metastasis of colorectal cancer cells. |
