| As a new resource food in 2012, the application fields of Panax ginseng are becoming more and more widely. Consumer have higher requirements to food than drug in taste. So it is often require additional sweeteners to enhance the mouthfeel during ginseng process. However, the effects of sugar water decoction on ginseng in quality evaluation and effectiveness have not been reported before. In the present study, the methods of HPLC, phenol-sulfuric acid and Folin-Ciocaltus were adopted respectively to determine the content of ginsenosides, soluble sugar content and the total phenolic content of sugar water decoction ginseng(SWG), water decoction ginseng(WG) and raw gisneng(RG) at first; Secondly, the methods of DPPH, FRAP were adopted respectively to determine the antioxidant activity of samples and the method of MTT was adopted to determine the effects of sugar water decoction on ginseng in inhibition proliferation of human breast cancer MCF-7 cell line cultured in vitro and screened out the better SWG preliminarily according to the antioxidant and antitumor activity; Thirdly, the study developed a new granule using fructose water decoction ginseng as material named “ginseng granule†and optimized the processing technology; Finally, the research further studied the antitumor activity in vivo using H22 tumor-bearing mice model and explored the mechanism of action. Experimental results are as follows:1. Ginsenosides F1, F2, Rg3, Compound K and the total content of 16 ginsenosides of ginseng increased significantly after processed with sugars(P<0.05). The content of soluble sugar and total polyphenol of SWG were significantly higher than RG as well as WG except for xylitol water decoction ginseng and the groups of glucose, maltose, sucrose, brown sugar, aspartame and chondroitin sulfate had higher content of total polyphenol than WG(P<0.05).2. Compared with RG and WG, sugar water decoction ginseng had higher DPPH radical scavenging capacity and FRAP(P<0.05).3. Sugar water decoction ginseng had higher inhibition proliferation activity to human breast cancer MCF-7 cell line than RG as well as WG(P<0.05). At the same time, fructose, maltose and chondroitin sulfate water decoction ginseng had the strongest inhibition proliferation activity to human breast cancer MCF-7 cell line among monosaccharide, disaccharide and polysaccharide, respectively.4. The optimal processing technology of “ginseng granule†using fructose as condiment: ginseng powder 50 g, water 250 mL, fructose dosage 13 g, processing temperature 100℃ and processing time 2 h. The product shows yellowish-brown and has uniformed color and luster, soft herbal fragrance, reasonable sweet taste and a good taste.5. Ginseng granule could inhibit the tumor growth of H22 tumor-bearing mice, increase the immune viscera index, protect the spleen tissue, induce the tumor tissue of H22 tumor-bearing mice to die, promote the apoptosis of H22 tumor cells, enhance the lymphocyte proliferation activity of spleen, improve the ratio of CD4+/CD8+, increase the levels of cytokine TNF-α, IFN-γ, IL-2, IL-6, IL-12 in serum and improve the content of immunoglobulin IgG, IgM in tumor-bearing mice(P<0.05). Although the positive drug 5-FU could inhibit the tumor growth of H22 tumor-bearing mice significantly, it also damaged the immune system of mice through decreasing the immune viscera index and the lymphocyte proliferation activity(P<0.05). |
