Objective The study was aimed to investigate the effects of Sijunzi decoction on the glucose absorption and the jejunum mucosa repair in spleen deficiency rats to investigate its mechanism of replenishing Qi and invigorating the spleen.Methods(1) Spleen deficiency model was prepared by reserpine via subcutaneous administration and was validated by general states and D-xylose excretory rate of rats.(2) The SD rats were divided randomly into 9 groups: normal control group, natural convalescence group, low dose spermidine group, medial dose spermidine group, high dose spermidine group, low dose Sijunzi Decoction group, medial dose Sijunzi Decoction group, high dose Sijunzi Decoction group and epidermal growth factor(EGF) control group. Combined in situ single-pass intestinal perfusion model, sulfuric acid-phenol method and UV were used to determine the glucose absorption. The jejunum mucosa pathological section was used to observe and compare the injury and repair of jejunum tissues of different groups.(3) The immunohistochemistry technology was used to determine the expression of SGLT-1 and GLUT-2 in jejunum tissues of different groups. The RT-PCR was used to analyze the difference of SGLT-1 m RNA and GLUT-2 m RNA in jejunum tissues of different groups.(4) The pre-column derivatization reversed-phase high performance liquid chromatography(HPLC) method was established and was used to determine the contents of polyamine in jejunum tissues of different groups. The real-time quantitative PCR(RT-PCR) was used to analyze the difference of ODC m RNA in jejunum tissues of different groups.Results(1) Compared with normal group, the body weight and D-xylose excretory rate of model group decreased significantly after reserpine injection(P<0.05).(2) Compared with normal control group, the glucose absorption significantly decreased and the jejunum mucosa significantly injured in natural convalescence group. Compared with natural convalescence group, the Ka and Papp of glucose was significantly increased and the injury of jejunum mucosa was significantly repaired in rats of spermidine groups, Sijunzi Decoction groups and EGF group(P<0.05).(3) Compared with natural convalescence group, the expression of SGLT-1 and GLUT-2 were increased in other treatment groups(P<0.05). The expression of SGLT-1 m RNA and GLUT-2 m RNA were increased significantly in other treatment groups(P<0.05).(4) Compared with natural convalescence group, the contents of spermidine, spermine and the expression of ODC m RNA were increased of spermidine groups, Sijunzi Decoction groups and EGF group(P<0.05).Conclusions Sijunzi Decoction, one of the most representative decoctions of replenishing Qi and invigorating the spleen, promoted the jejunum mucosa repair and the absorption of glucose in spleen deficiency rats. Its mechanism may facilitate the polyamine synthesis of intestinal mucosa to repair the jejunum mucosa, increasing the expression of SGLT-1 and GLUT-2.
