Objective:To analyze serum macrophage migration inhibitory factor(MIF) levels and MIF-rs755622 gene polymorphism in Shanxi population of patients with Graves disease,and explore its relationship with GD.Methods:Choose 60 patients with untreated GD as the case group( male 12 cases, female 48cases)) and 40 cases of healthy physical examination as the healthy control group(male 10 cases, female 30 cases). Serum macrophage migration inhibitory factor levels were detected by enzyme linked immune sorbent experiment method(ELISIA). The genotypes for the tested MIF-rs755622 gene polymorphisms in 60 GD patients and 40 normal controls were detected by polymerase chain reaction-restrictive fragment length polymorphism( PCR-RFLP).Results:1. The levels of serum MIF in patients with GD were 5625.20± 2403.13pg/ml which was obviously higher than that in healthy control group of 4651.11± 2266.55pg/ml, the difference was statistical significant in the two groups(t = 2.03, P = 0.045).2. In GD group, GG, GC and CC frequencies of MIF-rs755622 were 41.7%, 35%,23.3%, and G, C allele frequencies were 67.9%, 32.1%, respectively. In healthy controls,the corresponding genotype frequencies were 67.5%, 20.0%, 12.5% and G, C allele frequencies were 77.5%, 22.5%.MIF-rs755622 allele and genotype frequencies distribution were different in the two groups, the difference was statistical significant(P<0.05).3. The risk analysis of MIF-rs755622 gene polymorphism in GD showed that compared with GG, CC OR = 3.024(95% CI =0.951-9.617), P>0.05, there was no statistical difference in the two genotypes; GG OR=2.835( 95%CI = 1.065-7.549),P<0.05, the difference was statistical significant. The research indicated that individuals with GC genotype carrying the risk of suffering from GD are 4.355 times as much as that of the GG genotype crowd.4. The serum MIF level had significant difference among three different genotypes GG, GC, CC(F=7.763, P=0.001).Conclusion:1. The high expression of MIF in patient with GD which indicated its may be associated with the onset of GD.2. MIF-rs755622 GC gene polymorphism may be associated with the onset of GD, C allele may be one of the susceptibility genes of GD.3. MIF-rs755622 may be associated with the serum MIF level, if exploring the exact relationship MIF-rs755622 single nucleotide polymorphisms with GD has yet to enlarge sample to research in multicenter and many sites.
