Objective: 1. Study the changes of the ultrastructure in the damage area of the brain of the rats. In other words, by observing the changes of the cell nucleus and the mitochondria of the injury neurons to explore the inflammatory response of rats brain after traumatic brain injury. 2. By studying the distribution and the content of the NOGO- A in the damage area of rats brain after traumatic brain injury to explore the mechanism of the nerve regeneration of the central nervous system.Method: 1. Choose SD rats by Feeney method according to the principle of free fall injury to make the model traumatic brain injury. For traumatic brain injury model of rats with the same concentration fashudier tail intravenous injection. Put to death each group of rats(including control group, traumatic group and intervention group) after traumatic brain injury 24 hours.Using electron microscope to observe the changes of ultrastructure of the damage area and the surrounding tissue in each group, using the technique of immunohistochemistry to observe the distribution and the content of the NOGO- A in the damage area and the surrounding tissues. 2. Using SPSS19.0 software to process the statistical data. Using one-way analysis of variance to compare the date of each of groups.Results: 1. Electron microscopy shows: The control group and traumatic group:the injure of cell nucleus、Mitochondria and cells edema appear in the damage area of brains of rats of the traumatic group after traumatic brain injury 24 hours.The control group and intervention group: the injure of cell nucleus、Mitochondria and cells edema appear in the damage area of brains of rats of the intervention group after traumatic brain injury 24 hours. The traumatic group and intervention group: The damage degree of the traumatic group serious than intervention group. 2. Immunohistochemistry technique shows: The control group and traumatic group: there are differences in the distribution and the content of the NOGO- A in the damage area of rats brain of the control group and traumatic group(P<0.05). The trauma group content is higher than the control group. The control group and intervention group: there are differences in the distribution and the content of the NOGO- A in the damage area of rats brain of the control group and intervention group(P<0.05). The intervention group content is higher than the control group. The traumatic group and intervention group: there are differences in the distribution and the content of the NOGO- A in the damage area of rats brain of traumatic group and intervention group(P<0.05). The traumatic group content is higher than the intervention group.Conclusions: The injure of cell nucleus、Mitochondria and cells edema appear in the damage area of brains of rats after traumatic brain injury 24 hours. The distribution and the content of the NOGO- A in the damage area of rats have increased after traumatic brain injury 24 hours The RHO/ROCK signaling pathway was blocked can improve the damage of the ultrastructure of the brain cell in the damage area and reduce the content and distribution of NOGO-A in the damage area. It could promote the nerve regeneration of the central nervous system.
