Objective:To explore the value of diagnosis and monitoring the response to therapy by detectingplasma miR-193a-5p expression levels of AML patients in different phase of disease and normal controls. Method:(1) Peripheral blood samples were obtained from AML patients enrolled in hematology department of the Second Hospital of Shanxi Medical University from Jul 2015 to Dec 2015, including 40 de novo AML patients, 13 patients in CR and 10 patients in relapse. Peripheral blood samples from 15 healthy people were used as control.(2) Plasma miR-193a-5p expression levels were investigated using qRT-PCR(Quantitative Real-time PCR, qRT-PCR) and build the ROC curve. The nonparametricMann-Whitney U test was used. Results:(1) The plasma miR-193a-5p expression levels of AML patients group were obviously lower than that of normal control group(P<0.05);(2)The AUC of the ROC curve is 0.717, so the plasma miR-193a-5p is valuable in diagnosis of AML;(3)The plasma mi R-193a-5p expression levels of de novo and relapse AML groups were significantly lower than CR group and the control group(P<0.05), there was no significant difference in the expression level of the plasma miR-193a-5p between the de novo and relapse group(P > 0.05), also between the CR group and normalcontrol group(P > 0.05);(4) No significant correlation was found between the plasma miR-193a-5p level and age, gender, blast percentage of the bone marrow, peripheral blood leukocyte count, platelet count, hemoglobin,blast in BM,CD34 positive percentage, etc. Conclusion:The plasma miR-193a-5p expression level of AML patients is reduced. It may be used for evaluation of diagnosis and treatment in AML as a novel and noninvasive biomarker.
