Objective:To determine the human the relationship between the polymorphism of PARP-1,STAT4 gene and Graves’ disease(GD)in Han Group in shanxi area,aim to assess the pathogenesis of GD in the level of gene.Methods:The PARP-1 gene G1672 A alleles were typed by polymerase chain reaction based restriction fragment length polymorphism(PCR-RFLP)and The SNa Pshot single nucleotide polymorphism(SNP) genotyping method method respectively in 61 GD with male 16 and female 45(mean age 38.6±5.2)and 50 normal subjects with male1 13 and female37(mean age 38.6±5.2).The comparisons of genotype and allele frequencies were made between GD patients and normal group.Results:1.The distribution of the PARP-1 gene G1672 A and STAT4 gene rs7574865 in the control group and GD group did significantly devite from that expected for a Hardy-Weinberg equilibrium.2.Between the two groups of age,gender has no statistical significance.(p>0.05)3.The genetype frequencies of the PARP-1G1672 AGG,GA,AA in GD were55.5%,34.4%,9%,the main was GG genetype.The genetype frequencies of the PARP-1G1672 A in control case GG,GA,AA were 38%,54%,4%,the main was GAgenetype.The GG geneotype could significantly modify the 2.3 risk of GD.(p=0.04 CI1.034-5.122).The G and A allele frequencies were 72.9% and 27.1% in GD.The G and A allele frequencies were 65% and 35% in GD.The G allele could significantly modify the1.452 risk of GD.(P=0.02 CI 0.819-2.576)4.The genetype frequencies of the STAT4rs7574865 GG,GT,TT in GD were42.6%,24.5%,32.7%.The genetype frequencies of the STAT4rs7574865 GG,GT,TT in control case were 44.0%,44.0%,12%. There was no statistically significant difference between the two group.The G and A allele frequencies were 45.1% and 54.9% in GD.The G and T allele frequencies were 32.0% and68.0% in GD.The T allele could significantly modify the 1.744 risk of GD.(P=0.047 CI 1.005-3.027)Conclusion:1.PARP- 1G1672 A gene polymorphism may has something to do with the onset of GD in shanxi region, GG genotype is significantly higher than other groups of prevalence.And carry G gene increased the GD disease risk.2.PARP-1 G1672 A gene does not exist interaction with smoking effect.3.STAT4rs7574865 gene single nucleotide polymorphisms increase the risk of GD. |
