Objectives:To discusse the different expression levels of vitamin D receptor (VDR) and adipose differentiation related genes, including CCAAT enhancer binding protein alpha (C/EBPα), peroxisome proliferator activated receptor gamma (PPARγ) and sterol regulatory element binding protein-1c (SREBP-1c) mRNA in subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) of normal body mass index(BMI) persons and overweight and obesity persons,and to provide the pathophysiology theory basis of the occurrence of different types of obesity;and to lay the foundation of the mechanism of obesity patients prone to vitamin D deficiency and its prevention and control strategy according to the different expression levels of VDR in different parts of the adipose tissues and in different BMI groups.Methods:50 patients with open abdominal surgeries were choosen in this study, they were divided into two groups according to their BMIs:normal BMI group (group A,30 cases,18.5kg/m2≤BMI<24kg/m2), overweight and obesity group (group B,20 cases, BMI≥24 kg/m2). The RNA of SAT and VAT of all objects were extracted, and the general clinical data and biochemical data, such as total protein (TP), albumin (ALB), globulin (GLB), alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine (Cr), blood urea nitrogen (BUN) of all objects were measured, the expression levels of VDR, C/EBPα,PPARγ, and SREBP-1c mRNA in SAT and VAT of all subjects were detected; the different levels of the general clinical data and blood biochemical data between group A and B were compared with each other, and the different expression levels of the four genes in SAT and VAT in the same group of subjects were compared with each other and the different expression levels of the four genes in different parts of the adipose tissues were compared with each other respectively and the correlation between the four genes and the rest of the indicators were compared with each other respectively.Results:1ã€The expression levels of VDR,C/EBPa,PPARy and SREBP-lc mRNA between group A and B had no statistical significance in SAT (P>0.05); The expression levels of VDR,C/EBPa,PPARy and SREBP-lc mRNA between group A and B had no statistical significance in VAT (P>0.05).2ã€The expression levels of VDR mRNA in SAT of the objects in group A were higher than in VAT (P<0.05);and the expression levels of PPARy and SREBP-lc mRNA in SAT of the objects in group B were lower than in VAT (P<0.05).3ã€The single factor analysis among the expression levels of VDR, C/EBPa,PPARy, SREBP-lc mRNA and the rest of the indicators in SAT indicated that:(1) the expression levels of VDR,PPARy and SREBP-lc mRNA were significantly positively correlated with each other in SAT (P<0.01);(2) the expression levels of VDRmRNA were negatively correlated with ALP and FPG in SAT (P<0.05);(3) the expression levels of C/EBPa mRNA were negatively correlated with Na (P<0.05), and were significantly negatively correlated with Cl (P<0.05).4ã€The single factor analysis among the expression levels of VDR, C/EBPa,PPARy, SREBP-lc mRNA and the rest of the indicators in VAT indicated that:(1) the expression levels of VDR mRNA were positively correlated with SREBP-lc mRNA in VAT (P<0.05);(2) the expression levels of C/EBPa mRNA were positively correlated with PPARy mRNA (P<0.05);(3) the expression levels of PPARy mRNA were significantly positively correlated with SREBP-lc(P<0.01);(4) the expression levels of VDR mRNA were significantly negatively correlated with UA (P<0.01).Conclusions:1ã€The expression levels of VDR mRNA in SAT may has inhibitory effect on the occurrence of obesity, and the expression levels of VDR mRNA in VAT may play a promoting role in the occurrence of obesity.2ã€The occurrence of obesity may be associated with the lower expression levels of VDRã€PPARγã€SREBP-1c mRNA in SAT and/or with the higher expression levels of VDRã€PPARγã€SREBP-1c mRNA in VAT and the expression levels of these three genes in SAT may have synergy in promoting the occurrence of obesity.3. The expression levels of VDR mRNA were negatively correlated with FPG in SAT, the lower expression levels of VDR mRNA in SAT may lead to the elevated levels of FPG and the occurrence of the sugar metabolic disorders.4ã€The expression levels of VDR mRNA were negatively correlated with UA in VAT, the lower expression levels of VDR mRNA in VAT may lead to the elevated levels of UA, prompting that the lower expression levels of VDR mRNA in VAT may be correlated with the occurrence of hyperuricemia. |
