Objective (s):Lung cancer is the most common malignant tumors with high incidence and mortality, but the mechanism of lung cancer is unclear. Research have shown that the expression level of MGST1 gradually increased during the development of lung cancer, but the function of MGST1 in lung cancer is unclear. We examine the expression of MGST1 gene in non-small cell lung cancer cell lines and investigate the proliferation, apoptosis effect and cisplatin-sensitive of MGST1. The aim of this study was to investigate the physiological functions of MGST1, and to provide reliable basis for its potential as a therapeutic target for NSCLC.Methods:Western blot was applied to detect the expression level of MGST1 in NSCLC cell lines(A549. SPC-A-1、H129、NCI-H1581、NCI-H2405s SW900). The shRNA lentiviral expression vector targeting MGST1 gene was transfected in A549 cell line, the expression of MGST1 protein were detected by western blotting. The proliferation activity of A549 cells transfected with MGST1-shRNA and Scr-shRNA were determined by MTS assay. The colony-formation ability of A549 cells transfected with MGST1-shRNA and Scr-shRNA were determined by plate colony assay. The cell cycle and apoptosis rate of A549 cells transfected with MGST1-shRNA and Scr-shRNA were determined by flow cytometry method. Cisplatin inhibition rate of A549 cells transfected with MGST1-shRNA and Scr-shRNA were determined by MTS assay.Results:1. MGST1 is expressed in lung cancer cells generally. ● Western blotting results showed that MGST1 is expressed in A549, SPC-A-1, H1299, H1581, H2405, SW900, lung cancer cells.2. MGST1 promote the the proliferative ability of lung cancer cells. ● The results of the MTS assay showed that the proliferative ability presented a decrease after targeting MGST1 gene on A549. ● The condition of MGST1 gene silence, the colony forming ability of shMGST1 group was in than control group.3. MGST1 can inhibit the apoptosis of the lung cancer. ● The FCM analysis showed that the apoptosis rate increased after targeted MGST1 gene.4. MGST1 reduce sensitivity of lung caner cells to the cisplatin cytotoxicity ● The inhibition rate of shMGST1 group by cisplatin is higher than control group.Conclusion(s):MGST1 gene plays an important promote role in cell proliferation, clone formation ability, apoptotic resistance and reduce cisplatin sensitivity of NSCLC cells.
