| Ojective:Liver fibrosis is characterized by a variety of pathogenic factors caused by intrahepatic connective tissue hyperplasia, results in a diffuse Extracellular Matrix (ECM) in the liver of the pathological process of excessive precipitation. Hepatic Stellate cells (HSC) activation is thought to be the central link of liver fibrosis in the whole process is of great significance. This topic is to study the effect of Yanggan Auping mixture in reversing the progress of liver fibrosis and the mechanism of action. Using the rat liver fibrosis model induced by CCL4/olive oil, from the serum biochemical indexes, liver SOD, MDA, HyPcontent and liver tissue pathologic change to evaluate the role of Yanggan Auping treatment of liver fibrosis; At the same time, to further study the mechanism of action and the relationship between liver fibrosis and expression of cytokines TGF-beta1, TIMP-1.Methods:This paper is divided into two parts, theoretical and experimental research. Theory research, on the basis of ancient and modern literatures recorded in the system, records the understanding of traditional Chinese medicine for hepatic fibrosis and summarizes its etiology and pathogenesis, and about the development of the situation of resistance to the treatment of liver fibrosis. At the same time, it learns more about the research progress on the pathogenesis and treatment of liver fibrosis, providing theoretical basis for experimental study. Experimental research, by using the rat liver fibrosis model induced by CCL4/olive oil and using optical microscopy, automatic biochemical analyzer, lipid oxide test, rapid immunohistochemistry and molecular biology technology, observed Yanggan Auping mixtures effect of treatment of liver fibrosis, from the liver pathology change, serum biochemical indexes, liver SOD, MDA, HyP content and explored how Yanggan Auping effects the regulatory role of TGF-beta 1,TIMP-1, for the clinical application of Yanggan Auping mixture and new drug development provides new ideas. Results:1.Evaluation of the results of modeling:Model of hepatic fibrosis is successful, animals weight decreased and pathological manifestations of liver cell degeneration and necrosis, hepatic cord irregular, which marked fibrosis.2.On liver pathology:Yanggan Auping mixture in high dose group only see periportal inflammation and the formation of expanded, very few collagen fibers, which is the most effective.the colchicine group followed. In the other grops,Liver fibrosis also improved to some extent (P<0.01), the difference was statistically significant.3. On Serum Biochemical Indexes:Yanggan Auping mixture in high and middle dose group can significantly reduce serum AST, ALT, ALP, GGT, TB, DB level (P<0.01), a significant effect; colchicine groupã€Fuzhenghuayu group can also significantly reduce the level of liver function there was statistically significant difference compared with the model group (P<0.05).4. On liver tissue SOD, MDA, HyP Content:Yanggan Auping mixture in highã€middle and low dose group significantly increased SOD activity(P<0.01); In MDA, HyP content level, Yanggan Auping mixture in high dose group is lowest (P<0.01), followed by colchicine group and Yanggan Auping mixture in low dose group (P<0.01), the difference was statistically significant.5. On liver tissue TGF-β1, TIMP-1 expression:model group significantly increased the expression of TGF-β1, portal area, fibrous septa seen a lot of positive staining deepened. Yanggan Auping mixture in high dose group decreased TGF-β1, TIMP-1 deposition significantly(P<0.01), colchicine group and the FZHY group can reduced to some extent degrees TGF-β1, TIMP-1 level, the difference was statistically significant.Conclusions:This experiment using Yanggan Auping mixture on treatment of rat liver fibrosis model induced by CCL4/olive oil get the following conclusions:1. Yanggan Auping mixture can effectively inhibit the liver collagen fiber hyperplasia and extracellular matrix deposition, relieve inflammation of the liver cells, degeneration and necrosis, protect the structure of hepatic lobule, thus slowing down and reversing the progress of the liver fiber.2. Yanggan Aupng mixture can significantly reduce the level of serum biochemical indexes like ALT, AST, ALP, GGT, TB, DB, to reduce liver inflammation, protect the liver cells, play a significant curative effect.3. Yanggan Auping mixture has obvious effct of anti-lipid peroxidation and promoting the liver cell repairment, reducing the content of HyP, which inhibits the generation of collagen to slow down the liver fibrosis.4. In the development of liver fibrosis, Yanggan Auping mixture can inhibit the activation of HSC proliferation by reducing the secretion of TGF-beta 1 and the degradation of ECM such as collagen type I-. III by Inhibiting the expression of TIMP-1 and increasing the activity of MMP-1 to slow or even reverse hepatic fibrosis. |
