| Objective:Observing costimulatory molecular CD137 and CD28 expressed in CHB patients of liver tissue, to analyze the correlation of costimulatory molecular expression and CHB patients liver immune pathological damage degree, further explore CD28/B7 and CD137/CD137 L costimulatory signal to participate in the regulation mechanism of immune dysfunction disorder of the liver tissue in patients with CHB.Methods:1.The liver tissue collected by percutaneous liver puncture, grouped according to the liver HE staining results; 2. Detect each group ALT and TBIL liver function damage related indicators in peripheral blood; 3. Using ELISA method to detect the expression of IFN-γ and IL-10 and CD137 L level in peripheral blood; 4. Immunohistochemical method to detect each group of CD137 and CD28 expressed in the liver tissue; 5. Immunofluorescence method to detect each group of CD137 expression in liver tissue; 6. Western blot method to detect each group of CD137 and CD28 expressed in the liver tissue on the protein levels; 7. Fluorescence quantitative PCR method to detect each group of CD137 and CD28 expressed in the liver tissue on the m RNA levels.Results:1. According to HE staining of CHB patients with liver biopsy tissue pathology classification, it can be divided into mild CHB group, moderate CHB group and severe CHB group; 2. Healthy control group, mild CHB group, moderate CHB group and severe CHB group in the expression of ALT is on the rise, and compare differences between groups was statistically significant(P<0.05); there was no statistically significant difference between the mild CHB group and healthy control group in the expression of TBIL(P>0.05), but in moderate CHB group and severe CHB group the expression of TBIL increased obviously, the difference was statistically significant(P<0.05); 3. IL-10 in healthy control group, mild CHB group, moderate CHB group and severe CHB group expression is on the rise, there was statistically significant difference between the groups; IFN-γ in healthy control group compared with the expression level of CHB group, the difference was statistically significant(P<0.05); in mild CHB group,moderate CHB group and severe CHB group expression is on the decline, and the difference among three groups was statistically significant(P<0.05); 4. the expression of serum CD137 L in mild CHB group and moderate CHB group compared with healthy controls, there was no statistically significant difference(P<0.05); severe CHB group serum CD137 L expression compared with the above three groups, the difference had statistical significance(P<0.05); 5. Expression level of liver tissue CD28 m RNA in moderate CHB group and severe CHB group were higher than healthy control group and mild CHB group, the difference was statistically significant(P<0.05); 6. Expression level of liver tissue CD137 m RNA in severe CHB group was higher than healthy control group, mild CHB group and moderate CHB group, the difference was statistically significant(P<0.05); 7. The expression changes in protein levels of CD137 and CD28 were showed similar changes with the expression of mRNA levels.Conclusions: 1. In the liver tissues of CHB patients, the costimulatory molecular CD137 and CD28 expression level rising, has correlation with the immune pathological damage degree; 2. In the peripheral blood of CHB patients, the expression level of cytokine IL-10 rising and IFN-γ reducing, has correlation with the immune pathological damage degree; 3. The expression of costimulatory molecular CD137 and CD28, involved in regulating the liver tissue immune pathological injury of CHB patients. |
