Objective: To investigate the expression of ADAM29 and FAM135B in esophageal intraepithelial neoplasia and esophageal squamous cell carcinoma, and to explore the role of them in the development process. And study of expression with m RNAADAM29 and m RNA FAM135 B of esophageal squamous cell carcinoma before and after radiotherapy in peripheral blood.Methods: We used immunohistochemical(Envision) method to detect the expression of ADAM29 and FAM135 B in 40 cases of normal esophageal mucosa samples, 20 cases of mild intraepithelial neoplasia,20 cases of severe intraepithelial neoplasia and40 cases of esophageal squamous cell carcinomas(ESCC). At the same time,Application fluorescence quantitative RT- PCR( Reverse transcription polymerase chainr eaction) to detecte the expression changes of m RNAADAM29 and m RNAFAM135 B of 37 patients with esophageal carcinoma before and after radiotherapy in peripheral blood lymphocytes.Results : Immunohistochemical : 1.The positive expression rate of ADAM29 was17.5.0% in normal esophageal tissue, 60% in mild intraepithelial.neoplasia tissue, 90%in severe intraepithelial neoplasia tissue and 100% in ESCC tissue.The positive expression rate of FAM135 B was 25.0% in normal esophageal tissue, 55% in mild intraepithelial neoplasia tissue, 90% in severe intraepithelial neoplasia tissue and 100%in ESCC tissue. The positive expression of ADAM29 and FAM135 B in esophageal squamous cell carcinoma rates were significantly higher than the normal esophageal epithelium and esophageal Intraepithelial neoplasia(P<0.05).The positive expression of ADAM29 and FAM135 B in severe intraepithelial neoplasia tissue rates were higher than the normal esophageal epithelium and esophageal Intraepithelial neoplasia (P<0.05).2.The positive expression level of ADAM29 and FAM135 B are related to whether the lymph node metastasis and clinical stages and pathological stage(P<0.05),but not relevant to age and tumor size(P>0.05). 3.A positive correlation was found between ADAM29 overexpression and FAM135 B overexpression in esophageal lesion(P<0.05).RT-PCR : 1.The expression of ADAM29 on37 cases of esophageal cancer patients before radiation inperipheral mononuclear cells was 578.88 ± 1288.4342 times higher than that ofpatientswith esophageal carcinoma after radiotherapy(t=2.728,P=0.010)( P < 0.05).2.The expression of FAM135 B in esophageal cancer patients before radiation inperipheral mononuclear cells was 73.03±149.66479 times higher than that ofpatientswith esophageal carcinoma after radiotherapy(t=2.932,P=0.006)(P<0.05).Conclusion : 1.The expressions of ADAM29 and FAM135 B associated with the progress of esophageal cancer,whose expression can be used as early esophageal cancer diagnosis and provide the basis to guide the individualized treatment.2.The changes expression of m RNAADAM29 of esophageal squamous cell carcinoma before and after radiotherapy in the peripheral blood mononuclear cell have statistical significance( p < 0.05).Radiotherapy may have no effect on the expression of m RNAADAM29 in esophageal squamous cell carcinoma,Its expression may be used as evaluation criteria for the effect of radiotherapy.The changes expression of m RNAAFAM123 B of esophageal squamous cell carcinoma before and after radiotherapy in the peripheral blood mononuclear cell have no statistical significance( p> 0.05).Radiotherapy may have no effect on the expression of m RNAFAM135 B inesophageal squamous cell carcinoma,Its expression may not be used as evaluation criteria for the effect of radiotherapy.
