Objective: To investigate the effect of hydrogen rich saline on acute gouty arthritis model in rats, we detected the certain indicators:behavior score of the subject ankle joint and changes in joint circumference, the content of serum IL-1β, TNF-α, the pathological changes of articular synovia. Thus, we can explore the therapeutic effects of hydrogen rich saline on rats gouty arthritis, and providing a new method for the treatment of gouty arthritis. Methods: 40 rats were randomly divided into four groups. Control group,model group,hydrogen rich saline group and colchicine group(n=10). Colchicine group: treated with0.3KG colchicine orally, hydrogen rich saline group: were administrated of10 ml hydrogen rich saline continuously for 4 weeks, twice a day. The control group and model group were given the same volume of Twain-saline, rats acute gouty arthritis models were established on the last time after intragastric administration.Methods of modeling: Choose the right ankle joint lateral rear puncture point, use the iodophor disinfect the region, take 4 or 5 size syringe needle, make the mouth of needle inclined towards front which make a angle of4.5 degrees with shin bone, then inject into MSU solution 50 ul which concentration is 20mg/ml, take joint capsule on the side up as a standard.Control group were injected with the same volume of saline solvent-Twain. Theperimeter of right jiont was measured 24 hours before and after model establishment. After 72 hours of model establishment, blood samples were collected to test the level of Interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), and take the joint soft tissue to histopathology examination. Besides,combine the behaviors of rats to evaluate the therapeutic effect. Results: 1.changes in joint circumference: before model establishment, the perimete was no significant difference between each group(P>0.05);24 hours after model establishment, the ankles swelled to a peak, compared to the control group, the perimete of each group were significantly higher(P<0.01);and after drug treatment, compared to the model group, the Colchicine group and hydrogen rich saline group were all decreased with different leve, there was significantly difference between these groups. 2. Gait changes: 24 hours after model established, we observed the gait and make a score.Compared to the control group, model group had a significant difference in the gait(P<0.01), which indicated that the model was successful. Compared to the model group,Colchicine group and hydrogen rich saline group were all significantly getting better(P<0.01), there also have a significant difference between the groups. 3.The change of cytokines: 72 hours after model establishment, use the ELISA method to test the expression level of cytokines in serum. Compared to control group, serum IL-1β and TNF-α of model group was obviously increased, there was significantly difference betewen two groups(P<0.01); Compared to model group, serum IL-1β and TNF-α of model group was obviously decreased; but compared to Colchicine group, hydrogen rich saline group was no significantly difference betewen two groups(P<0.01). 4. Histopathology changes: the inflammatory reaction of subject ankle joint colchicine group and hydrogen rich saline group were improved than the model group. Conclusions: hydrogen richsaline can narrow the perimete of subjetc joint, improve the gait of rats, reduce the Inflammatory factors level and the inflammation of synovial. 2. hydrogen rich saline can effectively inhibit the gout arthritis of rats.
