| Objective: This paper used three different interventions on obese rats induced by high fat diet, which were aerobic exercise, composite plant polysaccharide gavage, aerobic exercise combined with composite plant polysaccharide gavage, to observe and analyze the effects of different interventions on obesity rats’ body weight and body fat content, and their play in the regulation of blood lipids, serum levels of inflammatory factors and the intestinal flora. Then analyze the possible mechanism of these interventions in the process of decreasing weight and fat.Methods: There were 70 male SD rats for this experiment. Randomly select 10 of which as the normal control group, only feeding ordinary feed. The rest 60 of which were used for establishing obese rats model, with a high fat feed. After 4 weeks the obesity model successfully built. Then divided the obesity rats into control group, aerobic exercise intervention group, compound polysaccharide intervention group, aerobic exercise + compound polysaccharide intervention group. We respectively adopt moderate intensity treadmill training, compound polysaccharide lavage and the combination of the above to interven the three different groups. Measure and record the morphology index of rats such as weigh and length, determine the change of rats’ serum lipid(TGã€TCã€LDLã€HDL) and inflammatory factors levels(IL-1ã€IL-6ã€TNF-αã€NF-κB) before and after intervention by using the method of ELISA, using 16 S rDNA sequencing to analyse and compare the difference of rats’ intestinal flora structure and change.Results:(1)The weight of obese rats induced by high-fat diet increased significantly(P<0.001). The serum TG, TC and LDL levels increased significantly(P<0.001), the serum HDL level decreased significantly(P<0.001). The serum inflammatory factors like IL-6, IL-1,TNF-α,NF-κB significantly increased(P<0.001). The Ruminococcaceae of obesity rats’ intestinal increased significantly, Clostridiaceae, Turicibacteraceae significantly reduced(P<0.05);the Chao index decreased significantly(P<0.05).(2)Aerobics intervention can make the weight and Lee index of obese rats decrease significantly(P<0.001). Aerobic exercise can regulate lipid metabolism effectively(P<0.001). The serum inflammatory factors significantly decreased(P<0.001). The Alcaligenaceae, Bifidobacteriaceae, Erysipelotrichaceae, Verrucomicrobiaceae of rats’ intestinal increased significantly, Helicobacteraceae and Ruminococcaceae significantly reduced(P<0.05).(3)Compound polysaccharide intervention can make the weight of obesity rats decrease significantly(P<0.05). It also can regulate lipid metabolism effectively(P<0.001). The serum inflammatory factors like IL-6, IL-1, TNF-α decreased significantly(P<0.05). The Bifidobacteriaceae, Clostridiaceae, Erysipelotrichaceae, Veillonellaceae, Turicibacteraceae of rats’ intestinal increased significantly, Desulforibrionaceae, Helicobacteraceae, Lactobacillaceae, Ruminococcaceae significantly reduced(P<0.05).(4)Aerobics + compound polysaccharide intervention can make the weight and Lee index of obese rats decrease significantly(P<0.001). It can make the serum TC level of obese rats decrease significantly(P<0.05). The Bifidobacteriaceae, Clostridiaceae, Erysipelotrichaceae of rats’ intestinal increased significantly(P<0.05).Conclusion:(1)By feeding high fat feed 4 weeks, we successfully established obese rats’ model.(2)A high-fat diet can induce the weight and body fat of obese rats significantly increased, synchronous with blood lipid metabolic abnormalities, inflammatory reaction aggravate, the species abundance, diversity and complexity of intestinal flora decreased.(3)By adjusting the composition of the intestinal flora to alleviate inflammatory response, effectively regulate lipid metabolism, thus aerobic exercise can achieve the purpose of weight and fat loss.(4)Complex plant polysaccharides can exert anti-inflammatory effects and effectively improve blood lipids by increasing the intestinal flora species relative abundance, while reducing weight.(5)Aerobic exercise + complex plant polysaccharides intervention can significantly reduce body weight and body fat of rats, its effect on blood lipid regulation and anti-inflammatory was no remarkable as the two separate intervention, but it not only can increase the relative abundance and diversity of the intestinal flora species, and can promote the complexity of animals’ intestinal flora. |
