Objectives:To study the trend of remifentanil and Ly294002 on the proliferation ability, migration ability and apoptosis ability of human hepatocellular carcinoma HepG2 cells, and the expression in hepatocellular carcinoma HepG2 cells of PTEN、AKT2 and mTOR mRNA.Methods:HepG2 cells were incubated with 100umol/L remifentanil,25 umol/L Ly294002 and 100umol/L remifentanil with 25 umol/L Ly294002. MTT method was used to detect HepG2 cell proliferation activity when HepG2 cells were incubated for 48h. We used the wound healing assay to detect the migration activity of HepG2 cells. And we used the flow cytometry instrument to detect the apoptosis and cell cycle of hepatocellular carcinoma HepG2 cells. The expression of PTEN、AKT2 and mTOR mRNA was detected by using quantitative realtime-PCR (RT-PCR).Results:1. The result from the MTT experiment testing human hepatocellular carcinoma HepG2 cells proliferation, compared with the absorbance of blank control group, the absorbance of R, L and R+L group decreased obviously, the differences were statistically significant (P <0.05); And R+L group compared with R and L group, the absorbance was more decreasing(P< 0.05).2. The healing rate in R, L and R+L group,was (19.42±1.92)%、(13.25±1.48)%、和(7.55±1.08)%,lower than that of the control group (36.25±4.20%); And R+L group compared with R and L group, migration ability was more decreasing (P <0.05).3. R, L and R+L group of cells in G0/G1 phase ratio were (70.91+1.43)%, (75.58+3.16)% and (80.39+1.17)%, higher than the blank control group (57.19+1.93)%(P<0.05); R, L and R+cells apoptosis rate were higher than the blank control group (P< 0.05); and R+L group compared with R and L group, the proportion of G0/G1 phase and apoptosis rate was more higher (P< 0.05).4. The PTEN mRNA expression in hepatocellular carcinoma HepG2 cells of R and L groups were relatively higher than control group (p< 0.05); and R group mTOR mRNA expression was higher than the control group, compared to the rest of the group, the mTOR mRNA expression in control group was lower (p<0.05); compared to control group, the AKT2 mRNA expression in the experimental group were lowered (p< 0.05).Conclusion:Remifentanil and Ly294002 can inhibit the proliferation ability and migration ability of human hepatocellular carcinoma HepG2 cells, and promote the apoptosis of human hepatocellular carcinoma HepG2 cells, and its mechanism may be associated with blocking human hepatocellular carcinoma HepG2 cells in G0/G1 phase, and adjusting the AKT2, PTEN and mTOR gene expression in liver cancer HepG2 cell; Ly294002 can enhance the influence of remifentanil on inhibiting the proliferation ability and migration ability of human hepatocellular carcinoma HepG2 cells, and promoting the apoptosis of human hepatocellular carcinoma HepG2 cells.
