[Objective] To investigate the relationship between Lysyl oxidase(LOX)and the development of pancreatic cancer, and the role of LOX in the recurrence and metastasis of pancreatic cancer after operation.[Methods] The expression of LOX was detected in pancreatic cancer cells, pancreatic cancer tissues and adjacent tissues of pancreatic cancer by RT-PCR and western blot. The invasion and migration of pancreatic cancer cell was detected by cell invasion and migration assay (Transwell). The clinical data of 64 patients with pancreatic cancer who underwent radical pancreatoduodenectomy were retrospectively analyzed. Moreover there were 22 cases of pancreatic cancer tissues, who have total follow-up data,were detected by immunohistochemistry, and the effect of LOX was analyzed on the recurrence and metastasis of pancreatic cancer after operation.[Results] 1.The expression of LOX mRNA in ASPC-1, PANC-1,SW1990 and BXPC-3 were difference, and the difference was statistically significant (F=733, P< 0.001; comparison enchother, P<0.001). The expression trend in four pancreatic cancer cell lines is ASPC-1>PANC-1>SW1990>BXPC-3.2.The expression of LOX protein in ASPC-1, PANC-1,SW1990 and BXPC-3 were difference, and the difference was statistically significant (F=6.021, P=0.019, comparison enchother, P<0.001).The expression trend in four pancreatic cancer cell lines is ASPC-1>PANC-1>SW 1990>BXPC-3.3.The invasion ability of LOX+ and LOX- which come from BXPC-3 cell was different, and the difference was statistically significant (t=-5.867, P<0.001). Furthermore the invasion ability trend is LOX-> LOX+.4.The survival rate and recurrence rate are different in the cases of 64 pancreatic cancer patients, which have different preoperative liver function, lymph node metastasis and postoperative pathological grading and staging,and patients who have diabetes or not. The difference was statistically significant (P<0.05);5.LOX protein was expressed in pancreatic cancer tissues and adjacent tissues of pancreatic cancer, and the positive rates were 31.8% and 36.3% respectively. There was no significant difference in survival rate and postoperative recurrence rate between LOX positive group and LOX negative group (P>0.05).[Conclusions] 1. Lysyl oxidase (LOX) were expressed in the four pancreatic cancer cell line with different invasive ability. With the cell invasive ability increased, LOX expression decreased. LOX may play an inhibitory role in the development process of pancreatic cancer.2. Preoperative liver function, lymph node metastasis, postoperative pathological grading and staging, and patients who have diabetes or not are important factors on influencing the prognosis of patients with pancreatic cancer.3. LOX protein was expressed in pancreatic cancer tissues and adjacent tissues of pancreatic cancer. There was no significant difference in survival rate and postoperative recurrence rate between LOX positive group and LOX negative group. The reason is that the samples is limited. |
