The Effect Of Qilougushen Decoction On Expression Of Nephrin And Podocin In Membranous Nephropathy Rate

Objective: the experiment evaluated the effect of Qi Lou Gu Shen deccction on rates with membranous nephropathyenal,including protein, urine serum biochemistry, renal pathologyin and the expression of nephrin mRNA and podocin mRNA. Exploring the mechanism of Qi Lou Gu Shen pills on the treatment of membranous nephropathy.Methods: Select 50 male Wister rats, random take 10 as blank group, the rest of rats was injected C-BSA on the tail vein every other day after a week of pre-immune to copy membranous nephropathy(MN) rats model.After the success of the building,the remaining rats were randomly divided into 4 groups,each group of 10,namely model group,low-dose group of Qi Lou Gu Shen medicine,high-dose group of Qi Lou Gu Shen medicine,tripterygium wilfordii control group.We have been watching 24 hours urinary protein quantitation,serum albumin, total proteintotal,alanine aminotransferase,aspartate aminotransferase,variations for fifty-six daya, and detecting kidney pathological chang eswith light microscope,and electron microscope.The expression of Nephrin mRNA, podocin mRNA in the renal tissues were tested by Real-Time PCR.Results:1 Qi Lou Gu Shen decoction can improve the general condition of membranous nephropathy rats,reduce 24 hours urinary protein quantitative,increase total proteintotal and serum albumin. Comparing with model control group,the difference was statistically significant(P<0.05, P<0.01),and the best therapeutic effectiveness is low-dose group of Qi Lou Gu Shen medicine,but there has no effective treatment for the liver function.Among these groups there was no significant difference with each other(P>0.05);2 Qi Lou GuShen decoction can to a certain extent,lighten the renal pathological damage of MN rats, reduce the de-position of immune complex inhabit the thickening of basement membrane of rats; 3 The medicine of Qi Lou Gu Shen pills can revisethe expression of Nephrin mRNA、Podocin mRNA in renal tissue of me mbranous nephropathy rats upwards.Compared with model control group difference was statistically significant(P<0.01), the most obvious is the low-dosegroupof Qi Lou Gu Shen medicine.Comparison of high-dose group of Qi Lou Gu Shen medicine and tripterygium wilfordii controled group, there was no statistically significant difference(P>0.05).Conclusions: 1 Qi Lou Gu Shen decoction can decline urine protein, increase serum albuminou,and have an treatment effects on membranous nephropathy rats.2 Qi Lou Gu Shen decoction can to a certain extent,lighten the renal pa-thological damage of MN rats,delay the progress of renal pathology.3 Qi Lou Gu Shen decoction can increase the expression of Nephrin mRNA、Podocin mRNA in renal tissues of MN rats, protect podocytes, maintain podocyte in membrane structure and the integrity of the glomerular filtration barrier function, have the effect of treatment of membranous nephropathy.
