The best treatment of severe liver disease caused by a variety of factors have been proved to be orthotopic liver transplantation, but the lack of liver donors and a series of problems caused by immune rejection greatly limits the use of the procedure, while in the screening of liver-related drugs as primary hepatocyte cell is difficult to culture and easy to mutate in the training process. And at the same times,with the depth of bone marrow mesenchymal stem cell research, more and more evidence indicate that bone marrow mesenchymal stem cells have potential to differentiate into hepatocyte-like cells. Thus, hepatocyte-like cell differentiated from bone marrow mesenchymal stem cells has good prospects in the field of regenerative medicine and drug screening.But for tree shrews, which is a new laboratory animals, there were no reserch reports for hepatogenic differentiation.To differentiate tree shrews’ bone marrow mesenchymal stem cells into hepatocyte-like cells, we isolated bone marrow mesenchymal stem cells with high purity and activity from the bone marrow of newborn tree shrews and obtained cell is verified by adipogenic and osteogenic differentiation and also improve the tree shrews marrow mesenchymal stem cells culture method which has been reported. At same time,a full-length coding sequence of HGF is cloned from the tree shrews liver tissue and its molecular characteristics is analyzed,which show a result that HGF of tree shrew and human share a highly similarity.Based on foundments above, we successfully differentiate tree shrews bone marrow mesenchymal stem cells into hepatocyte-like cells by use a claasical three-steps hepatogenic differentiation method (EGF, bFGF; bFGF, HGF, nicotinamide; OSM, Dex, ITS+premix), which provide a theoretical basis for the reserch of tree shrews in the field of stem cell transplantation therapy for liver disease and drug screening... |
