Cell growth was measured by MTT assay. Immunocytochemistry was carried out to detect the expression of VEGFR-2 and VEGFR-3. Transwell cell insert was uesd to compare the the effect of novel tyrosine kinase targeted inhibitor on the adhesion, invasion and migration of SCC-9.Results:MTT showed that compared with the blank control group, different concentrations of new drug and positive control group significantly inhibited the growth of SCC-9 (P<0.05), which cell growth inhibition rate rose along with the increase of drug concentration.The growth inhibition rate of new drug was higher than the positive control group. Immunocytochemistry demonstrated that different concentrations of new drug and positive control group both down-regulated the the expression of VEGFR-2 and VEGFR-3(P<0.05). Compared with the high expression of VEGFR-2 and VEGFR-3 on the cell membrane of scc-9 in blank control group, the degree of down-regulation increased with the drug concentration.The expression in the new drug was lower than the positive control group. Transwell cell insert experiment revealed that compared with the blank control, the number of penetrated cells in the new drug and positive control group decreased apparently(P<0.05), and reduced with the increase of drug concentration.The penetrated cells of the new drug group was less than the positive control group.Conclusion:The novel tyrosine kinase targeted inhibitor exhibite a significant inhibitory effect on the growth of SCC-9, down-regulate the expression of VEGFR-2 and VEGFR-3 of SCC-9, and it can obviously inhibit the invasion and migration ability of SCC-9, and the inhibited effect may enhance with the increasing of the concentration of the new drug.
