| Objective:Establish the adriamycin-induced myocardial injury model in rats,and treated by traditional Chinese medicine decoction Yiqi Huoxue Prescription to intervention.To study the effects of Yiqi Huoxue Prescription on the superoxide dismutase(SOD),malondialdehyde(MDA),creatine kinase isoenzyme(CK-MB) and aspartate aminotransferase(AST) in rats with adriamycin-induced myocardial inj ury.Discuss the mechanism of the protective effect of Yiqi Huoxue Prescription on adriamycin-induced myocardial injury in rat with free radical injury.Methods:1. Grouping and Administration:Forty Wistar rats were randomly divided into 4groups,with 10 rats in each group,including Blank group, driamycin group(A DM group), adriamycin+traditional Chinese medicine low dose group(Low dos e group), adriamycin+traditional Chinese medicine high dose group(High dose group). blank group were treated with distilled water intragastric administration2ml/only, while treated with physiological saline intraperitoneal injection 2ml/o nly, once every two days, a total of 6 times; Except the Blank group, the othe r groups were given intraperitoneal injection of 2ml ADM(2.5mg/kg), once ev ery two days, to establish the adriamycin-induced myocardial injury model in r ats,meanwhile,ADM group were treated with distilled water intragastric admi nistration 2ml/only, once every two days; Low dose group and H igh dose gro up were treated with Yiqi Huoxue Prescription(10.5g/kg;5.25g/kg) intragastric a dministration 2ml/only, once every two days, a total of 6 times.2. Indexes testing: Observe the changes of general situation of rats during the experiment; The body weight of rats was measured before each administration and before blood sampling. After the end of the experiment, the weight averag e change curves of the rats were drawn and the value of weight loss in rats were calculated. After the rats were killed, the heart was removed and weighedand calculated the HW/BW of rats. After the end of the experiment, test the SOD activity, MDA content in serum and the content of CK-MB and AST in cardiac muscle tissue by ELISA method. Collected the myocardial tissue of rat s, preparation of myocardial tissue pathological sections and observe the pathol ogical changes of myocardial tissue.Result:1. General state: Rats in each experimental group compared with the blank gro up, the survival status of rats was worse than that of the blank group, body w eight decreased significantly compared with the blank group and the HW/BW r atio ratio increased significantly. But compared with the ADM group, the living condition of each Yiqi Huoxue Prescription group improved significantly, weig ht loss reduced than ADM group(P<0.01), HW/BW ratio decreased than ADM group and the High dose group decreased is particularly evident(P<0.01).The effects of Yiqi Huoxue Prescription on improving the general living condition,slowing down the weight loss and reducing the degree of heart failure is obvi ous.2. ELISA method for detection of SOD, MDA, CK-MB, AST results show that:After administration, the SOD activity of ADM group was significantly lower t han that of the blank group(P<0.01), MDA, CK-MB and AST levels were sig nificantly higher than those in the blank group(P<0.01), and the difference ha ve statistical significance; The SOD activity was significantly higher(P<0.05 or P<0.01) in the treatment group than in the ADM group, and the effect of the High dose group was the most significant. Compared with ADM group, the lev els of MDA, AST and CK-MB in Low dose group and High dose group were significantly lower than that of ADM group(P<0.01 or P<0.05).3. Pathological observation of cardiac muscle: In the blank group, the structure of the myocardium was normal, and there was no damage to the myocardial cells. The myocardial cell damage was most obvious in ADM group, and the s everity caused by ADM toxicity was the most serious.The myocardial tissue oft he two Yiqi Huoxue Prescription groups had different degree of injury, but thedegree of lesion was lower than that of the ADM group, the degree of High d ose group was significantly lower than that of Low dose group.Conclusion: Yiqi Huoxue Prescription has a protective effect on adriamycin ind uced myocardial injury in myocardial tissue of rats, It may be possible to impr ove the SOD content of rat to enhance the ability of myocardial cells to clear oxygen free radicals, inhibit lipid peroxidation of myocardial cell membrane so as to reduce the generation of the end product MDA, protect the structure an d function of myocardial cell membrane and organelles, reduce the release of AST and CK-MB caused by myocardial cell injury. In short, Yiqi Huoxue Pres cription can effectively eliminate oxygen free radicals and play an antioxidant r ole, ensure the normal dosage of adriamycin, and effectively prevent and cure the toxic damage to the heart of adriamycin. |
