The node of lotus rhizome(Oujie) is the dry rhizome section of Nelumbo nucifera Gaertn. It is a section between two lotus knots, which main active component is polyphenols. Polyphenols have strong antioxidant activity that prevent cell damage by scavenging free radicals. Oujie is a good antioxidant raw materials, but it often be disposed as waste during handling and processing of the lotus products, resources can not be effectively utilized. To improve the efficient use of wasted Oujie, and developing related products, it will contribute to the efficient use of Oujie.The experiment were developed by extract Oujie, preparation of Oujie oral liquid and the test of polyphenols, and optimize extract technology and preparation technology by amplification test, and obtain a more ideal production process. The results and conclusions of this study are as follows:During the preparation of Oujie extract liquid, the present experiment by selecting its extraction solvent, solid-liquid ratio, the choice of ultrasonic time and ultrasonic power, to determine the final choice of Oujie extract: ethanol concentration of 35%(V / V), solid-liquid ratio 1:25, ultrasonic power 120 W, ultrasonic time 40 min. At the same time through amplification test, the extraction technology of Oujie extract was accessed.During the preparation of Oujie oral liquid, the present experiment take taste as the standard by the amount of sucrose and citric acid screening, and ultimately determine the proportion of sucrose was added to 25%(W / V), citric acid of 1.25%(W / V). In the amplification test, we got the best preparation technology of Oujie Oral Liquid.During the determination of the polyphenol content of Oujie extract, the present experiment determine the amount by 10% Folin-phenol solution and 7.5% sodium carbonate solution volume screening test and the volume were 3ml and 6ml respectively. Then linearity, repeatability and recovery of this method was validated, the results show, gallic acid reference substance linear is good between 23.80~119.0μg, regression equation was Y = 0.0044 X + 0.0041, correlation coefficient is 0.9994, repeatability of RSD = 1.5%(n = 6), recovery rate were: 98.5%, 98.9%, 99.1%, 99.8%, 98.2% and 100.3%, indicate the test method of the polyphenol content is suitable of Oujie extract. |
