Purpose:By studying the hypoxic exposure on one-off skeletal muscle of rats after eccentric exercise Calpain- 3 and the change of dysferlin to explore whether hypoxic exposure through the inhibition of p94 and dysferlin exacerbating membrane injury prevents injury recovery.Methods: Select 70 eight weeks rats, divided into C,N24,N48,N72,H24,H48 and H72, except the C group, the othe group go for one-off training exercise after the adaptive training,put the rats into the oxygen enviroment and hypoxiaenviroment, then take back after 24 hours, 48 hours and 72 hours, measured the p94 and dysferlin protein expression and gene m RNA expression of the rats’ gastrocnemius muscle, take each two EBD injection to observe the cell membrane permeability change. Use the SPSS19.0 softwareto make the measured data processing and analysis.Results: After one-off eccentric exercise, the m RNA level of p94 increased until 24 hours, then come down. The different between N24 and NC0 N,N48,N72 is significant(P<0.05). There is significantly different between N24 and H24(P<0.01). After one-off eccentric exercise, the m RNA level of dysferlin increased until 48 hours, then come down. There is significantly different between N48 and N72, H48 and H72(P<0.01).There is no significantly different in the protein level.Conclusions: 1.The mainly effect of p94 and dysferlin on the membrane repair mechanism is in the 48 hours after one-off eccentric exercise;2. The effect of p94 and dysferlin on the membrane repair mechanism is not one of the the membrane repair mechanism by hypoxic exposure.
