| Objective: To study the effect of curcumin on NF-κB activation and the treatment outcome and mechanism of curcumin on polyethylene particle-induced osteolysis, providing new insights into the prevention and treatment of prosthetic aseptic loosening.Methods: Thirty 4 week-old male BALB/c mice were selected randomly and set up air pouch on back, killed ten of them randomly after one week and taken out the skull. The skull was implanted into the air pouch of other mice,which were divided into the following four groups: blank group(group A), polyethylene group(group B), solvent group(group C), curcumin group(group D). 1ml saline was injected in the air pouch of blank group and the same amout of polyethylene particles suspension was injected in the rest three groups. Curcumin was dissolved in DMSO and corn oil was added till DMSO volume fraction reached 5% to prepare curcumin solution, meanwhlie, control solution without curcumin was prepared. From day 9, 0.2ml curcumin solution(curcumin dosage50 mg/kg/Day) was injected into the air puch of curcumin group, 0.2ml control solution was injected into solventand the rest two groups was given the same aomout of saline every day till the 21 st day. Harvested all the mice and taken out air pouch as well as sterile. TRAP staining was used to detect the amount of osteoclast in skull. Extracted total RNA and total protein after air pouch liguid nitrogen grinding, Q-PCR was performed to measure the mRNA level of c-FOSã€NFATc1; westen blot was performed to test the protein level of IκBα and p-IκBα in air pouch tissue.Result:The TRAP staining result revealed: dyed osteoclast in polyethylene and solvent group were more than the other two groups, however, blank and curcumin group showed little or no osteoclast dyeing. QPCR: the mRNA level of c-FOS: polyethylene group >solvent group >blank group >curcumin group,former two groups compared with latter two,the difference between each group was statistically significant(P < 0.01). The mRNA level of NFATc1: polyethylene group >solvent group >blank group >curcumin group, former two groups compared with latter two,the difference between each group was statistically significant(P < 0.01). Western blot: the IκBα protein grey value: curcumin group >blank group>solvent group >polyethylene group, former two groups compared with latter two, the difference between each group was statistically significant(P < 0.01). p-IκBα protein grey value: polyethylene group > solvent group >blank group> curcumin group, former two groups compared with latter two,the difference between each group was statistically significant(P < 0.01).Conclusion:In mice air pouch bone graft model, curcumin could reduce ultra-high molecular weight polyethylene induced bone dissolution reaction by inhibiting NF-κB activation and c- FOSã€NFATc1 expression. |
