Objective: to observe Gubi mixture effect for IL-6 beta. TNF-alpha in the cut the double forelegs induce the rats to up joint Hulth method modle of the KOA knee osteoarthritis model in rats, while studying the degeneration of the cervical spine of the rat model.Methods: to copy a human development similar to osteoarthritis of knee joint double joint Hulth erect forelimbs model of rat osteoarthritis of the knee. Choosing the 15 th pregnancy age 12 in SD rats,and raised into different cage,putting pups born 10 th to 40 th day,Sorted by weight size were randomly divided into four groups,respectively for the blank control group, and normal saline group, and osaminethacine group, and Gubi mixture group, 10 rats in each group. Removing four group double arms by laparoscopic surgery in anesthesia,then raised in original rat cages,using Hulth method with ritht knee after four months. Gubi mixture group set by 0.5m L/100 g weight intragastric infusion with mixture, glucosamine indomethacin group given enteric-coated tablets 2mg/100 g in the same volume of distilled water and allow it to melt shake, Intragastriced administration of saline group by 0.5ml/100 g weight given saline, the control group without any intervention, intervention procedures Last 4 weeks, then rats will be killed under anesthesia, removing the right knee and cervical spine, then fixed, decalcification, dehydration, embedding,then slicing section,fishingsection, roasting section(overnight), then with HE staining, methyl aniline blue staining, safranin green staining, and Immunohistochemical staining,then observing and analyzing.Results: joint cartilage surface of right knee in the blank control group, and saline group are less smoother than the joint cartilage surface in osaminethacine group and Gubi mixture group in HE staining, methyl aniline blue staining, safranin green staining. The joint surfaces appear crack in the blank control and saline group. The cartilage cell distributions are not uniform, and not neatly, beside the gradation are fuzzy. Cartilage cells are in a cluster, and tide mark not full. Local staining not are uniform, even light staining and lost staining. IL-1 β and TNF-alpha percentage of expression of positive cells of Immunohistochemical staining in blank control group and normal saline group are higher than the last two groups.(2) Distribution of cartilage endplate cells of cervical spine in blank control group and normal saline group is less uniform than the last two groups. The cartilage endplate cells are not neatly and the gradation are fuzzy, even in a cluster. Local staining are not uniform, even light staining and lost staining. Fibrosus annulus are misarrange and fissured.Nucleus are collapsed, and little gas in the disc, called "vacuumphenomenon†Conclusions:(1) Gubi mixture has an effect on the cutting the double forelegs induce the rats to up with Hulth method modle of the KOA knee osteoarthritis model, can delay apoptosis of cartilage cell, by lowering the expression of IL-1 β and TNF-alpha, delayed knee osteoarthritis chondrocytes apoptosis process.(2) the Gubi mixture also has an impact on the cervical spine of the rat model,delayed the process of degeneration of the cervical spine. |
