Recently years, the diabetes patient increased rapidly and threated to our health. Drugs aimed at diabetes commercially always have strong effect of decrease the blood-glucose, but they are not unsuitable for long-term use because of different degrees of side effects. Traditional Chinese medicine, which has multi-channel and multi-target comprehensive treatment advantages and little side effects, has been widely noted. The decreaseing effect of two Chinese medicinal materials propolis and honeysuckle on blood-glucose of alloxan-induced diabetic mice were observed. The fasting blood-glucose, glucose tolerance, body weight, liver weight, kidney weight of mice, the T-SOD, MDA, GSH-Px, AST, ALT, and liver T-SOD, MDA, GSH-Px of mice serum were detected to determine the optimal dose. Based on the optimal dose, the optimal compound of propolis and honeysuckle were investigated. The results were as follows:1. The optimal extraction parameters of honeysuckleDry Honeysuckle were crushed and extracted by ultrasonic assisted extraction. Ethanol concentration, solid-liquid ratio, ultrasonic power and ultrasonic time were designed to observe the yield. The orthogonal analysis were conducted for the optimized parameters.The results showed that the optimal parameters were:60% ethanol, solid-liquid ratio 1:10, ultrasonic power 150W and ultrasonic time 30min. The extraction yield was 44.69% under these conditions.2. The effect of honeysuckle extract on diabetic miceThe effect of honeysuckle extract on diabetic mice was determined for the optimal dose. The control group, model control group, positive control group and honeysuckle 0.5g/kg, lg/kg, 2g/kg,4g/kg,6g/kg dose group were designed. The results showed that:honeysuckle extract could improve all the physical and biochemical indicators of diabetic mice as well as syndrome of polydipsia, polyphagia, weight loss and other symptoms were greatly improved. The best dose of honeysuckle was 1g/kg.3. The effect of ethanol extract of propolis on diabetic miceThe effect of honeysuckle extract on diabetic mice was studied to determine the optimal dose. The effect of the compound of propolis and honeysuckle on diabetic miceThe effect of propolis and honeysuckle in different proportions were studied with control group, model control group, positive control group, honeysuckle treatment group (1g/kg,2g/kg), propolis treatment group (O.lg/kg,0.2g/kg), the compound of propolis and honeysuckle (orthogonal of propolis (O.lg/kg,0.2g/kg)and honeysuckle (lg/kg,2g/kg) treatment groups: group â… (honeysuckle 1g/kg+propolis 0.1g/kg), groupâ…¡ (honeysuckle 1g/kg+propolis 0.2g/kg), group â…¢(honeysuckle 2g/kg+propolis 0.1g/kg), group â…£(honeysuckle 2g/kg+propolis 0.2g/kg). The results showed that:groups used medicine could significantly improved the physical and biochemical indicators of diabetic mice as well as syndrome of polydipsia, polyphagia, weight loss and other symptoms. The treatment effect of compound of propolis and honeysuckle were superior than honeysuckle group and propolis group, respectively. The optimal compound group was 1g/kg honeysuckle and 0.2g/kg propolis, which displayed the strongest hypoglycemic effect. And the treatment effect of 0.2g/kg propolis in compounds is better than 0.1g/kg propolis in compounds. |
