Research On The Mechanism Of Oldenlandia Diffusa Willd Polysaccharide To Non-small Cell Lung Cancer A549

Hedyotis diffusa (HD) is the dry herb of Hedyotis that is the rubiaceae annual herbaceous plant, the determination of this research is to discuss the antitumor activity and mechanism of Hedyotis diffusa polysaccharide(HDP) by culture non-small cell lung cancer(NSLCC) A549 cell experiments,(1)Use CCK-8 to detect the effect of HDP on non-small cell lung cancer A549 proliferation with different concentrations and different time. (2)Use FCM, fluorescent staining, DNA Ladder and scratch experience to detect different concentrations HDP to A549 on cell cycle, apoptosis and transwell. (3)Use western blot to detect the effect of different concentrations HDP on protein expression of MMP-9、E-CAD and Caspase-3.(4) Take QPCR to discover the influence of different concentrations HDP on gens expression of Bcl、Survivin、CyclinD 1 and CDK4.Experiment Result Expresses, (1) HDP can inhibit the proliferation of A549 in vitro obviously. And it has time dependence and concentration dependence. (2)The results of FCM, fluorescent staining, DNA Ladder show that HDP can stop the cell cycle of A549 at G2 period, and can accelerate apoptosis obviously to A549.Scratch experience result shows that HDP can inhibit A549 migrating, and has concentration dependence.(3) HDP can obviously reduce the protein expression of MMP-9, and increase the protein expression of Caspase-3.This conclusion denominate HDP can inhibit degradation of protein components in cell basement membrane and extracellular matrix, and can promote cell adhesion strength within the organization, and then effectively inhibit A549 cell infiltrate and metastasis of A549 cell to surrounding tissues.HDP can accelerate A549 cell apoptosis, then effectively inhibit A549 proliferation.(4) HDP can obviously reduce the gens’transcription of Bcl-2 and Survivin. This conclusion point that the mechanism of HDP induce A549 apoptosis is HDP can inhibit transcription and gens’expression of Bcl-2 and Survivin, thus HDP play a role in antitumor. We also confirm HDP can accelerate A549 apoptosis by reduce the RNA expression of CyclinD 1 and CDK4, prevent the conformation of CyclinD-CDK complex, postpone the promoting and transformation of cell cycle in different period.Conclusion:HDP can inhibit cell proliferation, have antitumor activity, the mechanism may be HDP can promote A549 cell apoptosis, block the cell cycle of A549, and control invasion and metastasis, that is has obvious antitumor effect.
