Objective The expression of COX-2,MMP-14 and TRPP-2 were detected by immunohistochemical method in Hypopharyngeal squamous cell carcinoma tissues, and compared with the expression level of tissues adjacent to carcinoma.To analyze and examine the relationship between excessive expression of COX-2,MMP-14 and TRPP-2 and development, invasion,metastasis and prognosis of Hypopharyngeal cancer.Methods During January 2008 to January 2011, 50 cases of carcinoma tissues were collected from patients who were operated in the hospital.Meanwhile,50 cases of tissues adjacent to carcinoma were used for comparison.Immunohistochemical methods were used to detected the expression of COX-2,MMP-14 and TRPP-2 in Hypopharyngeal carcinoma tissues and tissues adjacent to carcinoma.The mean optical density of COX-2,MMP-14 and TRPP-2 were measured by Image-proplus 6.0 software.The SPSS software(version 17.0) was used to analyze the date.Results Immunohistochemistry showed that COX-2,MMP-14 and TRPP-2 protein expressed in 50 cases of carcinoma tissues were significantly higher than those in tissues adjacent to carcinoma(COX-2:t=14.310,p<0.001;MMP-14:t=18.617,p<0.001; TRPP-2:t=15.742,p<0.001).Besides,the expression of COX-2,MMP-14 and TRPP-2 in carcinoma tissues were associated with T classification(COX-2:t=-4.871,p<0.001; MMP-14:t=-3.810,p<0.001;TRPP-2:t=-3.599,p<0.001),clinical stage(COX-2:t=-6.891, p<0.001;MMP-14:t=-12,p<0.001;TRPP-2:t=-9.791,p<0.001),lymph node metastasis(COX-2:t=-6.591,p<0.001;MMP-14:t=-8.298,p<0.001;TRPP-2:t=-6.775,p<0.001;) and histological grades(COX-2:t=-5.196,p<0.001;MMP-14:t=-4.704,p<0.001;TRPP-2: t=-3.621,p<0.001;). What’s more, Kaplan-Meier method is used to analysis of the relationship between the expression of three kinds proteins and survival rate.Results show that survival time is correlate with the P50,P75,P80,P90 level of COX-2,MMP-14 and TRPP-2 expression.Univariate and multivariate Cox’s analysis indicates that recurrence ≧P75 level of TRPP-2 expression and ≧P90 level of TRPP-2 expression had an independent prognostic effect on prognosis.The protein expression of COX-2 was positively correlated with the protein expression of MMP-14 in Hypopharyngeal squamous cell carcinoma tissues.The protein expression of COX-2 was positively correlated with the protein expression of TRPP-2.The protein expression of MMP-14 was positively correlated with the protein expression of TRPP-2(r=0.688,p<0.001).Conclusion 1.COX-2,MMP-14 and TRPP-2 proteins expressed in carcinoma tissues were significantly higher than those in tissues adjacent to carcinoma.This suggests that COX-2,MMP-14 and TRPP-2 proteins play important roles in hypopharyngeal carcinoma. 2.The expression of COX-2,MMP-14 and TRPP-2 in carcinoma tissues were associated with T classification,clinical stage,lymph node metastasis and histological grades.This indicates that COX-2,MMP-14 and TRPP-2 proteins overexpression may be associated with tumor invasion and metastasis. 3.Survival analysis showed survival time is significantly correlated with of COX-2,MMP-14 and TRPP-2 overexpression, and suggests that the relationship between the level of COX-2,MMP-14 and TRPP-2 and prognosis. 4.COX-2 expression was positively correlated with MMP-14 expression in Hypopharyngeal squamous cell carcinoma tissues.COX-2 expression was positively correlated with TRPP-2 expression,also MMP-14 expression was positively correlated with TRPP-2 expression.It provides us a way to predict the progression and prognosis of the tumor.
