| Objective To investigate the relationship between the changes of manganese superoxide dismutase(MnSOD), sirtuin 3 (SIRT3) with adjuvant arthritis (AA) rats.Method Complete Freund’s adjuvant (FCA)of subcutaneous injection of SD rats induced by AA, the AA rats were put to death on the day 13,19 and 25, calculate the liver index, serum aspartate transaminase (AST), alanine aminotransferase (ALT) were detected, methane dicarboxylic aldehyde (MDA), glutathione peroxidase (GSH-px) and superoxide dismutase (SOD) in liver homogenate were also detected, and MnSOD, SIRT3 protein expression changes were detected in the liver tissue by immunohistochemistry and Western blot.Result Compared with the normal rats, arthritis model of SD rats induced by complete adjuvant on the day 3, performance as acute inflammation, red swelling of the paw and joints, the ROM of ankle joint decreased, activity limited, secondary inflammation development, AA rat model was successfully established. AA rats on the day 13, the liver index, serum transaminase eleations, liver homogenate MDA increased, GSH-px, SOD activity decreased (P<0.05or P<0.01).The expressions of MnSOD and SIRT3 protein decreased in liver tissue. Joint and liver HE staining of AA rats showed:Synovial tissue proliferation and edema, formation of pannus, small vascular proliferation in synovial tissue, infiltration of inflammatory cell, articular cartilage destruction and bone erosions. In the late time joint space narrowing, form irreversible joint change; structure of liver injuries, performance as infiltration of inflammatory cell, hepatic blood sinus congestion, hepatocytes point or slice necrosis.Conclusion There is liver damage in AA model group rats, oxidative stress index increases in liver homogenate.The mechanism is related to the expression of MnSOD and SIRT3 in the liver. |
