Immunohistochemical Research Of Connexin 43(Cx43) In Human Dental Pulp

Connexin protein 43(Cx43) directly involves in the regulation of gap junction intercellular communication to transfer the information and energy between cells.And then,it regulates a series of physiological and biochemical process,such as the metabolism proliferation and differentiation of cells.Cx43 plays an important role in the early vertebrate nervous system conduction, cardiovascular development, bone formation and mineralization.At the same time,it is closely related to the occurrence of diseases such as the cardiovascular system, nervous system, urogenital system and tumor development.In the recent years,some scholars studys have found that the expression of Cx43 protein in dental pulp tissue, but no study has found the function of Cx43 in inflammatory dental pulp.This experiment uses immunohistochemical method to discuss the expression and function of Cx43 protein in the process of human pulpitis disease preliminarily.Objective: To observe the expression and localization of connexin 43 in different state of human dental pulp tissue by immunohistochemical method.And To discuss the mechanism of Cx43 in human inflammatory dental pulp tissue preliminarily and to provide new way for the treatment of pulpitis.Method: The fresh clinical pulp tissue out of dental is fixed in 4% paraformaldehyde and paraffin embed routinely.We use Hematoxylin-eosin staining method to observe the pathology of pulp tissue.And the tissue is divided into three groups combining with clinical manifestations:the healthy group,the acute pulpitis group and the chronic pulpitis. The Image pro- plus software is used to analysis the average optical density of each picture, and the immunohistochemical staining is used to evaluate the expression of Cx43 in three groups of dental pulp tissue.Results: The result of HE staining shows vascular expansion congestion in the pulp lesion of acute or chronic inflammatory dental pulp, a large number of inflammatory cells swim out of blood vessels and tend to inflammatory center, such as neutrophils, lymphocytes and plasma cells.The Immunohistochemical results shows that Cx43 is expressed in three groups of dental pulp tissues, the positive cell density and yellow dye is significantly different in different areas and different specimens of dental pulp.Microscopically,The expression of Cx43 protein is obviously increased in inflammation group(P < 0.05),and the expression of Cx43 protein in Acute inflammation is stronger than chronic inflammation group(P < 0.05).Conclusions: Cx43 is expressed in human inflammatory dental pulp tissue, and plays an important role in the occurrence and development process of pulpitis.
