Combining clinical data with pathological data and applying statistics methodology, we carried out correlation analysis to explore the relationship between these variants and osteosarcoma differentiation, metastasis and malignancy.Method: All the patients of our study were selected from the Second Hospital of Jilin University. Bone tumor biopsies from April 2009 to January 2013 were collected and then diagnosed by two senior pathologists independently, combined with clinical information, to determine tumor type, degree of differentiation and metastasis. We collected a total of 23 cases patients of bone tumor, in which 10 patients were osteosarcoma: 7 males and 3 females, with age range(12-87 years old); six cases were poorly differentiated and four cases were well-differentiated; 6 cases with metastasis and 4 cases without metastasis. There were 8 patients with Giant cell tumor, in which 4 were males and 4 were females, with age range(26-54 years old). Osteochondroma in 5 cases, of which 3 were males and 2 were females, with age range(9-73 years old). Clinical and pathological information are complete, all patients did not receive radiotherapy and chemotherapy before operation. We divided the collected biopsies into group osteosarcoma, group giant cell tumor of bone and group osteochondroma, then extracted RNA and reverse transcribed into c DNA, designed q PCR primers and carried out quantitative real-time PCR. Calculating CT value of every gene in every sample with the use of software IDEA and takeing number 1 sample as the reference standard, we applied the method of 2- â–³ â–³ CT to calculate the relative expression value of every gene in every sample, with mean ± standard deviation( x ±s) to represent the relative expression value of each gene in every group, and finally carried out correlation analysis.Result:1.The expression of CD44 s in every group.The relative expression value of CD44 s in group osteosarcoma and group giant cell tumor of bone[(2.33 ± 1.38),(3.03 ± 1.92)] were higher than the group osteochondroma(0.36±0.30), the difference was statistically significant, P all <0.05; The relative expression value of CD44 s between group osteosarcoma and group giant cell tumor of bone was not different statistically significant, P> 0.05.2. The expression of CD44v6 in every group.The relative expression value of CD44v6 in group osteosarcoma and group giant cell tumor of bone[(0.48±0.41),(1.32±1.43)] were higher than the group osteochondroma(0.10±0.12), the difference was statistically significant, P all<0.05; The relative expression value of CD44v6 between group osteosarcoma and group giant cell tumor of bone was not different statistically significant, P> 0.05.3. The expression of CD44v9 in every group.The relative expression value of CD44v9 between group osteosarcoma(4.75±4.10)and group giant cell tumor of bone(2.70±2.43) was not different statistically significant, P>0.05; and CD44v9 shows no expression in group osteochondroma.4. The correlation between the expression of CD44 s, CD44v6 and CD44v9 in group osteosarcoma and differentiation of osteosarcoma.There was no significant difference of the relative expression value of CD44 s between poorly differentiated osteosarcoma samples(2.66±1.73) and well-differentiat-ed osteosarcoma samples(1.85 ± 0.52), P> 0.05;There was no significant difference of the relative expression value of CD44v6 between poorly differentiated osteosarcoma samples(0.42±0.42) and well-differentiat-ed osteosarcoma samples(0.55±0.45), P> 0.05;The relative expression value of CD44v9 in poorly differentiated osteosarcoma samples(6.90 ± 4.07) was higher than the well-differentiated osteosarcoma samples( 1.55 ± 0.99), the difference was statistically significant, P <0.05.5. The correlation between the expression of CD44 s, CD44v6 and CD44v9 in group osteosarcoma and metastasis of osteosarcoma.There was no significant difference of the relative expression value of CD44 s between metastasis osteosarcoma samples(2.66±1.73) and non-metastasis osteosarco-ma samples(1.84±0.51), P> 0.05;There was no significant difference of the relative expression value of CD44v6 between metastasis osteosarcoma samples(0.42±0.42) and non-metastasis osteosarco-ma samples(0.55±0.45), P> 0.05;The relative expression value of CD44v9 in metastasis osteosarcoma samples(6.90 ± 4.07) was higher than the non-metastasis osteosarcoma samples( 1.55 ± 0.99), the difference was statistically significant, P <0.05.Conclusion:1. The expression of CD44sã€CD44v6 and CD44v9 in different bone tumor are different : The expression of CD44 s and CD44v6 in osteosarcoma and giant cell tumor of bone were significantly higher than that in osteochondroma(P all<0.05), but there was no significant difference between osteosarcoma and giant cell tumor(P> 0.05); CD44v9 shows no expression in osteochondroma and there was no significant difference between osteosarcoma and giant cell tumor of bone(P> 0.05);2. Only the expression of CD44v9 has correlation with the differentiation and metastasis of osteosarcoma, whereas CD44 s and CD44v6 has no such correlation :The expression of CD44v9 in poorly differentiated osteosarcoma was higher than that in well-differentiated osteosarcoma(P<0.05) and the expression of CD44v9 in metastasis osteosarcoma was higher than that in non-metastasis osteosarcoma(P <0.05);3.
