| ObjectiveTo investigate the effect of cluster needling of scalp acupuncture combined with treadmill training on sciatic nerve motoring functional i ndex (SFI), Wet weight of triceps surae and expression of Murfl pr otein in rats with sciatic nerve injury, to verify the effectiveness of cluster needling of scalp acupuncture combined with treadmill training on delaying denervated muscle atrophy. Discuss the regulatory mechanis ms of cluster needling of scalp acupuncture combined with treadmill tra ining on denervated muscle atrophy.Methods84 male SD rats were randomly divided into 5 groups, blank group, sham group (n=6), model group, treadmill group, cluster needling of s calp acupuncture combined with treadmill group (n=24).Each group was further divided into four subgroups (n=6), including 7,14,21and 28 da ys subgroups. The sciatic nerve of rats was clamped in model group, tr eadmill group and cluster needling of scalp acupuncture combined with treadmill group. The rats in treadmill group were treated by treadmill t raining, and the rats in cluster needling of scalp acupuncture combined with treadmill group were treated by cluster needling of scalp acupun cture and treadmill training,while the rats in blank and sham and mo del groups were received no intervention. At each time point, the sciat ic nerve function index (SFI) and the wet weight of triceps surae were measured,the expression of Murfl was examined by Western Blottin-g.Results1.SFI:The SFI of blank and sham group showed the normal value 0, over time, the value of SFI in the other groups became larger.T he value of SFI of each group was the smallest in the early stage of i njury (7d),the SFI value of each group became the biggest at the time point 28d.At each time point, the value of SFI of the model group is the smallest, at each time point the value of SFI in treadmill group wa s higher than that in the model group, the difference was statistically s ignificant (P<0.05) and the value of SFI in the cluster needling of scal p acupuncture combined with treadmill group was higher than that in th e treadmill group, the difference was statistically significant (P<0.05).2. Wet weight ratio:The wet weight ratio of the blank group and the sham group was normal value-1. Over time,The wet weight ratio of other groups decreased gradually.The highest at 7d and the lowest at 28d among all the time points.Wet weight ratio showed maximum at 7d,at each time point,the wet weight ratio of the treadmill group was greater than that of the model group, the difference was statistic ally significant (P<0.05), at each time point,the wet weight ratio of th e cluster needling of scalp acupuncture combined with treadmill gro up was greater than that of the treadmill group, the difference was stati stically significant (P<0.05).3. Murfl protein:The expression of Murfl protein in the blank gro up and shame group showed little expression, the expression of Murfl protein in the other groups was highest at 7d and lowest at 14d,compa red with model group,the expression of Murfl protein was higher in th e treadmill group and the cluster needling of scalp acupuncture combine d with treadmill group, the difference was statistically significant (P<0. 05), at each time point, compared with the treadmill group,the expres sion of Murfl was lower in the scalp acupuncture combined with tread mill group,the difference was statistically significant (P<0.05)Conclusion1. The cluster needling of scalp acupuncture combined with treadmi 11 training can improve the value of SFI, slow down the decline rate of the wet weight ratio of triceps surae, to prevent the muscle atrophy and the cluster needling of scalp acupuncture combined with treadmill training was better than simple treadmill training.2. The cluster needling of scalp acupuncture combined with treadmi 11 training can delay denervated muscle atrophy, this may be related to decreasing the expression of Murfl. |
