| Objective: To build a BALB/c mouse model of endotoxemia with injection of LPS intraperitoneally,and to give intervention treatment with injection of remifentanil intraperitoneally.Accordingly,to research the role of remifentanil on apoptosis of thymocyte in mouse.Methods: By using the random number table method to divide 45 BALB/c mice into 3 groups.The normal control group(NS group,n=15): physiological saline was used for injection intraperitoneally. Lipopolysaccharide group(LPS group, n=15):8mg/kg LPS was injected intraperitoneally.Remifentanil treatment group(RE group,n=15):the method of LPS injection was the same as that in LPS group,and then gave 18μg/kg remifentanil immediately and intraperitoneally every 10 minutes,3 times in total.Observed the general condition of the 3 groups,measuring the levels of the interleukin-10(IL-10),tumor necrosis factor-α(TNF-α) in the serum,and the protein content of Bcl-2.Results: 1.The changes of general condition:Compared with the normal control group,LPS group mice showed eating less,horripilation,shaking,less activity,diarrhoea and so on.However, these symptoms were alleviated in Re group.2.The changes of TNF-α and IL-10 in mice serum:Compared with the normal control group,the levels of TNF-α and IL-10 in the LPS group were obviously increased(P<0.01).The levels of IL-10 and TNF-α in RE group were both lower than that in LPS group(P<0.05),but it was still higher than that in normal control group(P<0.05).3.The expression of Bcl-2 in thymus:Compared with the normal control group,the expression of Bcl-2 in LPS group was down-regulated(P<0.05).In RE group,the expression of Bcl-2 was up-regulated(P<0.05)by comparison with LPS group.Conclusion: Pretreatment of remifentanil has a protective effect on apoptosis of thymocyte in a mouse model of endotoxemia. |
