Objective:Through the observation of different concentrations of SANBI Decoction in Treating Adjuvant Induced AA model rat cytokine changes, and to observe the joint swelling and synovium, cartilage tissue pathological degree of injury and the mechanism of SANBI Decoction in the treatment of rheumatoid arthritis, for clinical provides reliable experimental basis.Methods:Model of rheumatoid arthritis in the right ankle joint of rats with adjuvant injection. Randomly divided into blank group, model group, the group of three, the high and middle dose group. Observation of AA group rats at different time point joint score; homemade foot volume measuring instrument to observe changes in joint volume of rats; ankle synovium, cartilage, bone pathological changes were observed under light microscopy; the content of enzyme linked immunosorbent assay (ELISA) to detect the serum levels of IL-6, IL-8, IL-10, IL-17, MCP-1; immunohisto chemical method to detect the synovial TNF-α, MCP-1, RANTES, TGF-β and IL-17 levels of expression; X-ray observation rats foot bone erosion.Results:1. Right to primary primary compared with model control group,3 weeks later, SANBI Decoction high dose group of foot joint inflammation score reduction was statistically significant (P<0.05), the 4 weeks of Tripterygium wilfordii control group and SANBI Decoction high, middle dose group of foot joint inflammation score reduction was statistically significant (P<0.05).2, compared with the blank group,AA group of rat paw volume at each time point were significantly increased (P<0.05);and model group compared SANBI Decoction high dose group, Tripterygium wilfordii group rats right foot volume in week 2 decreased significantly (P<0.05) and Tripterygium wilfordii group compared SANBI Decoction high, medium dose group foot volume change had no statistical significance (P>0.05).3, Compared with model group, the pathological changes of SANBI Decoction of high, middle dose group and the joint of the hook group were significantly reduced, and there was statistical significance (P<0.05). Compared with SANBI Decoction, the pathological changes of the synovial joints of the high and medium dose group were not significantly improved (P>0.05).4, compared with the blank group, more than AA group of rat IL-6,IL-10, MCP-1, IL-8, IL-17 levels significantly increased (P<0.05);and the model group compared SANBI Decoction high, middle dose group and twp group, IL-6, MCP-1, IL-8, IL-17 in decreased significantly, IL-10 levels were significantly increased (P<0.05);and Tripterygium wilfordii group compared SANBI Decoction high, middle dose group, IL-6, IL-10, MCP-1, IL-8, IL-17 content changes (P>0.05).5,cumulative optical density value comparison:compared with the blank control group, AA group rats TNF-aand MCP-1,RANTES,TGF-βand IL-17 value increased (P<0.05); compared with the model group, the three arthralgia, high dose group and Tripterygium wilfordii group TNF-α and MCP-1, RANTES,IL-17 values were significantly lower,TGF-βincreased significantly (P<0.05); and Tripterygium wilfordii group compared SANBI Decoction middle dose group of MCP-1 elevated values, TGF-p, IL-17 value decreased (P<0.05).6, X-ray observation of the original right foot found: SANBI Decoction, high dose group and the rats in the group of rats in the knee joint surrounding soft tissue swelling, no osteoporosis, bone destruction and new bone formation. Model group showed osteoporosis, cartilage destruction and extensive destruction of bone, while the rats in each group AA rats with secondary foot lesions were relatively light.Conclusion:1、SANBI decoction can significantly reduce the volume of AA rats, arthritis score, improve the level of activity in rats.2、Through the X-ray and light microscope observation of rats, the high and middle dose of SANBI decoction can obviously improve the pathological morphology, bone and cartilage damage in AA rats.3、SANBI decoction may regulate anti-inflammatory and proinflammatory cytokines by reducing serum levels of IL-6, IL-8, MCP-1, IL-17, and elevated anti-inflammatory cytokines IL-10 in serum of AA rats.4、SANBI decoction may inhibit the expression of IL-6, IL-8, TNF-α, RANTES, TGF-β, IL-17, and improve the secretion and activity of AA, inhibit the proliferation of synovial cells, and reduce the damage of the joints.
