Aim:Observe the adenosine receptor transduction in ventrolateral medulla of the acute myocardial ischemia rat regulated by acupuncture selecting acupoints on the involved meridian, investigate the probably central neural mechanism of the coronary artery disease (myocardial ischemic) treated by acupuncture.Methods:Choose healthy adult SD rats as the research obj ect, then randomly divide them into four groups, blank group, model group, acupuncture group, and sham operation group. By ligation the left anterior descending coronary artery (LADCA) to prepare the acute myocardial ischemic model. Rats in acupuncture group will treat by electroacupuncture after AMI model prepared, Using Hans-LH200, and the stimulating parameters:the needling mode,2/100 Hz frequency and 0.5±0.2 mA, as the rat limb shaking slightly, lasts for 30 minutes, one time a day lasts for five days. Observe the ST height and HRV by the II lead of ECG. The ischemic area of myocardium evaluated by 2,3,5-tripheyltetrazolium Chloride (TTC) stain. Hematoxylin eosin (HE) staining to observe the pathological changes of myocardial ischemia. Cardiomyocyte apoptosis is detected by Tunel-POD. The adenosine A2a receptor expression,the ERK1/2 expression and the pERK1/2 are detected by western blot.Result:1. The ST segment of rats elevate after ligated left anterior descending coronary artery during the operation of AMI model; Detect the II lead of ECG when complete the intervention, the ST segment both descreased in AMI model group and acupuncture group,but there is no diffeerence between the two groups (P>0.05);2. Stained by TTC,the color of normal myocardium is red,while the color of ischemic myocardium is white,but the ischemic area has no difference between AMI model group and acupuncture group (P>0.05)3. The LF/HF of HRV elevate after ligated left anterior descending coronary artery during the operation of AMI model. The LF/HF decreased in acupuncture group after complete the acupuncture intervention, compared with AMI model group (P< 0.05);4. The result of myocardium histomorphology shows that, in the AMI model group,the myocardial fiber is fracture and irregular, a large number of inflammatory cells infiltration, hyperplasia of connective tissue. In the acupuncture group, the myocardial fibers is just irregular, a small amount of inflammatory cell infiltration and hyperplasia of connective tissue5. The results of the myocardial cells apoptosis detected by Tunel-POD shows that, the rate of myocardial cells apoptosis in sham operation group has no significant difference with the blank group the rate of myocardial cells apoptosis in the acupuncture group is lower than the AMI model group (P< 0.05)6. The expression of adenosine A2a receptor and the ERK1/2,and the level of ERK1/2 phosphorylation in ventrolateral medulla of rats in AMI model group is higher than blank group (P< 0.05);The expression of adenosine A2a receptor and the ERK1/2,and the level of ERK1/2 phosphorylation in ventrolateral medulla of rats in acupuncture group is lower than AMI group (P< 0.05).Conclusion:1. Acupuncture the selecting acupoints along the meridian could relieve the myocardium ischemic condition of AMI model rats.2. The regulation of the heart autonomic nervous balance, and reduce rat myocardial cell apoptosis rate by acupuncture the selecting acupoints along the meridian, maybe the reason that the relieve of the myocardium ischemic condition of AMI model rats by acupuncture.3. adenosine signaling transduction in ventrolateral medulla of acute myocardial ischemia rat that is the potential mechanism that acupuncture treat the coronary artery disease.
