ObjectiveTo observe the effect of buyanghuanwu decoction in pulmonary fibrosis rats model caused by bleomycin,and to explore the mechanism of buyanghuanwu decoction on pulmonary fibrosis.Method144 SD rats(SPF),height 200±10g, were randomly divided into 6 groups:control group,BLM group,positive drugs group,experimental largeã€middleã€small group. Control group were established by injecting saline,the five groups were established by injection bleomycin(5mg/kg)into its trachea.Converting the buyanghuanwu decoction dosage between human and animals according to the concentration.large:middle:small=4:2:l.The concentration of the large dosage is 27.32g/kg, the middle is 13.66g/kg, the small is 6.83g/kg. The rats were treated with medicines after 24 hours. Control group and BLM group were intervened by 10ml/kg-d distilled water,prednisone group was given 4.2mg/kg-d prednisone solution,the large dosage of buyanghuanwu decoction group was given its solution by 27.32g/kg-d, the middle dosage of buyanghuanwu decoction group was given its solution by 13.66g/kg-d, the small dosage of buyanghuanwu decoction group was given its solution by 6.83g/kg-d.The six goups were put to death on drug intervention after 7ã€14ã€28d respectively.the BALF and lung tissue were colected.HE and Masson staining were used to observe the degree of alveolar inflammation and fibrosis in lung tissue, ELISA was applied for IL-6ã€IL-8 quantitation and immunohistochemical staining (S-P) was applied for expression of TβRI.Resultsl.Pathomorphology:The structure of lung tissue in control group rats is clear, no morphological changes;compared with the control group,the model group and each medicated groups were mainly featured with destroyed alveolar structure,Infiltration of inflammatory cells and deposition of collagen fibers on different level;compared with the model group,the medicated groups could improve the alveolitis and pulmonary fibrosis on pathomorphology in different time points (p<0.05 或 p< 0.01).the effect of reducing alveolitis and pulmonary fibrosis of buyanghuanwu decoction large-dose group had an advantage over that of the prednisone group on 28th day (p<0.05)2.General status:the rats of control group is robust, they had sensitive reaction, smooth fur, normal food intake,increased body weight. The rats in the model group had the phenomena of lack of energy,glazing vision,unresponsive, breathing breathlessness, lack of exercise, dry dull hair, less eating, slow increase weight. compared with the model group,otther medicated group’s had a good mentality, rapid response, stable breathing, shiny hair,the decline extent of activity and food intake had different degrees of improvement with time, increased gradually weight.3.The expression of IL-6:Compared with control group, IL-6 expression of BALF in each group rats were gradully increased,the model group was the most obvious(P< 0.01).Compared with model group,each medicated group had different decrease in the expression of IL-6(P< 0.05, or P<0.01).Compared with prednisone group,the expression of IL-6 of buyanghuanwu decoction large-dose gorup obviously decrease on the 28th day(P<0.05).4.The expression of IL-8:Compared with control group, IL-8 expression of BALF in each group rats were gradully increased,the model group was the most obvious(P< 0.01).Compared with model group,each medicated group had different decrease in the expression of IL-8(P< 0.05, or P<0.01).Compared with prednisone group,the expression of IL-8 of buyanghuanwu decoction large-dose gorup obviously decrease on the 14th and 28th day(P< 0.05),as well as that of buyanghuanwu decoction middle-dose gorup on the 28th (P<0.05).5.The expression of TβR I:In the lung tissue of rats,the positive expression of TβRI was mainly distributed in the lung interstitium and around blood vessels. The expression of TβR I in control group on the 7th,14th and 28th day was small.There was no significant difference between the groups. Compared with control group, the expression of TβR I in the model group increased in different time points,the 28th was the most obvious.Compared with model group,each medicated group had different decrease in the expression of TβR â… , the 14th and 28th were the most obvious(P< 0.01).Compared with prednisone group,the expression of TβR â… of buyanghuanwu decoction large-dose gorup obviously decrease on the 14th and 28th day,as well as that of buyanghuanwu decoction middle-dose gorup on the 28th (P<0.05).Conclusion1.In different time points,the degree of alveolar inflammation and pulmonary fibrosis in lung tissue of model group rats were significantly higher than that of control group, and the pathological morphological changes were consistent with the literature.The PF rat model induced by BLM was successfully duplicated.2.Buyanghuanwu decoction could significantly reduce the alveolar inflammation and delay the formation of pulmonary fibrosis,its role was similar to prednisone.3.The large dose of Buyanghuanwu decoction could inhibition the expression of IL-6, IL-8, TβR â… ,its effect was better than the other groups. So the large dose of Buyanghuanwu decoction had a good concentration-response relationship on the prevention and cure of pulmonary fibrosis, it may be suitable for clinical application and dosage of prevention and cure.4. Buyanghuanwu decoction can improve alveolar inflammation by inhibiting cytokines expression of IL-6, IL-8 in IPF rat BALF; By inhibiting the expression of TβR â… ,reducing the activity of TGF-β1, thus reducing the degree of pulmonary fibrosis. |
