Ischemic stroke have a strong impact on people’s health and daily life with its high morbidity, high mortality and disability rate. How to prevent and control stroke disease has attracted a widely attention. Xingnaojing Injection was an effective and rare clinical adverse reactions traditional Chinese medicine injection on treating stroke which was consisted of four herbs included four flavor herbs radix curcumae, musk, borneol and gardenia. It show outstanding curative effect on cerebrovascular disease, alcoholism, expecially the ischemic stroke. We analysed the mainly contents of XNJI by chirality gas chromatography, founding that it was consist of L-Camphor, D-camphor, L-borneol, D-borneol, iso-borneol, and musk. Then we focus on screening the effective ingredient by pharmacological model in vivo and in vitro, wanna to developed a new repurified XNJI which would be stable and effective for better quality control and security.Objective(1) Screening the efficacy ingredients of XNJI on ischemic stroke model in vitro.(2) Screening the efficacy ingredients of XNJI on ischemic stroke model in vivo.(3) Seeking an efficacy refined Xingnaojing Injection(RXNJI) by optimizating the percentage of the three active ingredients (L-camphor, D-borneol, musk ketone).Methods(1) Serum free induced SH-SY5H cells injury model, H202 induced HT-22 cells injury model, and glutamic acid induced cortex neurons injury model were applied to screen the medicable constituents of XNJI.(2) Unilateral carotid artery occlusion with nervi vagus ligation mice were applied to screen the medicable constituents of XNJI.(3) Uniform design was used to obtain the reasonable compatibility of XNJI conbined with the experimental data of middle cerebral artery occlusion on rat.Results(1) The viability of serum free induced SH-SY5Y cells injury was improved by L-camphor(80 μM、120 μM、160 μM), D-borneol(30μM、50μM,70μM) and iso- borneol(15μM、25μM、35μM).(2) The viability of H2O2 induced HT-22 cells injury was improved by L-camphor(150μM、100μM、50μM), D-borneol(30μM、50μM、70μM),iso-borneol(15μM、25μM、35μM),and musk ketone(10μM).(3) The viability of glumate acid induced cortex neurons injury was improved by D-borneol (33μM)、musk ketone (0.1μM)(4) L- camphor, L-borneol and D-borneol reduced the nerve function index of unilateral carotid artery occlusion with nervi vagus ligation mice, While L- camphor, L-borneol, D- borneol and musk relieving the cerebral water content of operative mice.(5) RXNJ reduced the nerve function index of unilateral carotid artery occlusion with nervi vagus ligation mice and middle cerebral artery occlusion (MCAO) rat, reduced the mortality and infarction area ratio on MCAO rat.ConclusionThe composition of RXNJ was consist of L-camphor, borneol and musk ketone as (8:7:1, M/M/M) and (5:3:2, M/M/M).
