This paper optimized the anthemis nobilis and indocalamus latifolius leaves essential oil supercritical extraction technology, steam distillation essential oil extraction process and extraction process of flavonoids, and the essential oils and flavonoids were identified. The main research contents and conclusions are as follows:The orthogonal test was used to optimize the Anthemis nobilis essential oil and indocalamus latifolius leaves essential oil supercritical extraction process. Anthemis nobilis essential oil supercritical extraction rate of the primary and secondary order pressure > extraction temperature > extraction time. Anthemis nobilis essential oil supercritical extraction process for optimal dynamic extraction time of 3 hours, extraction pressure of 40 MPa, and extraction temperature at 50 degrees. According to the optimization design of orthogonal experiment, the supercritical fluid extraction of Anthemis nobilis essential oil, essential oil yield was 6.8%. The effect of the order of indocalamus latifolius leaves essential oil: extraction time > extraction pressure > extraction temperature. Extraction temperature at 40 degrees. Under this condition, indocalamus latifolius leaves essential oil yield was 1.66%.Using single factor method to optimize the technology of preparation of indocalamus latifolius leaves anthemis nobilis essential oil and essential oil by steam distillation method. Anthemis nobilis essential oil extraction time 5h, ratio of material to liquid 1:30, under these conditions the oil yield is 0.53%.Chromatography were identified Rome Chamomile essential oil and supercritical steam distillation of essential oil by comprehensive two dimensional gas. The highest content of 10 kinds of compounds included 6 kinds of esters, 2 kinds of ketones, 1 organic acids and 1 alkanes. The water vapor content in the essential oil of ten highest compounds included 7 kinds of esters, alkanes, ketones and alcohols were 1.By comprehensive two-dimensional gas chromatography were identified from Indocalamus leaves supercritical oil and steam distillation. Supercritical fluid extraction of essential oil were identified 88 kinds of compounds containing more than 0.02% of the peak area, the five peak area of more than 1% compounds respectively is hexadecanoic acid, Cyclohexene, 1-methyl-4-(1-methylethenyl)-,(S)-, 9,12-Tetradecadien-1-ol,(Z,E)-, 2-Hexenal, Steam distillation of essential oil were identified 74 kinds of compounds, Cyclohexene, 1-methyl-4-(1-methylethenyl)-,(S)-peak area of up to 13.56%. Steam distillation of essential oil of indocalamus latifolius leaves, the highest content of compounds respectively is Cyclohexene, 1-methyl-4-(1-methylethenyl)-(S)- hexadecanoic acid 9,12-Tetradecadien-1-ol,(Z,E)-,2-Hexenal, Cholest-5-en-3-ol(3á), 3,7,11,15-Tetramethyl-2-hexadecen-1-ol.Optimization of extraction process of flavonoids from anthemis nobilis and indocalamus latifolius leaves. The optimum extraction conditions of anthemis nobilis as the alcohol concentration of 80%, solid-liquid ratio 1:30, extraction time 2h. Under this condition, the flavonoids extraction rate was 6.1% in anthemis nobilis. Indocalamus latifolius leaves flavonoids optimum extraction conditions were ethanol concentration 60%, solid-liquid ratio 1:20, extraction time 1.5h. Under these conditions, the yield of flavonoids was 1.35% of indocalamus latifolius leaves.Anthemis nobilis and indocalamus latifolius leaves flavonoids were identified by HPLC-DAD method. Anthemis nobilis identified 22 compounds. Included 7 kinds of quinic acid, 15 kinds of flavonoids. 3-methoxyoxaloyl-1, 5-dicaffeoylquinic, apigenin, apigenin-7-Orutinoside, 1,4-di-caffeoylquinic acid were highest. Identified 4 kinds of flavonoids in indocalamus latifolius leaves. They were 6-C-glucosyl-8-C-arabinosy-chrysin, 6,8-C,C-diglucolylapigeninã€7-O-glucolylchrysinã€orientin. By adding 10 μL/cigarette of anthemis nobilis essential oil and indocalamus latifolius leaves essential oil can reduce the stimulation, improve the quality of aroma respectively. By adding 30 μg/cigarette of anthemis nobilis and indocalamus latifolius leaves flavonoids can improve the dry feeling of cigarette smoke and soft the smoke respectively. |
