Due to the progress of technology on deep cryopreservation, it becomes possible to preserve the living tissue and cells, even the human life. In the past several tens years, based on the principle of freezing turning into cell damage, the deep cryopreservation technology has made lot of achievements on freezing methods, cryoprotectants, the tank for cryopreservation and recovery and elution, which leads its application in medical treatment and cell resources storage. Here begining from the advance of the deep cryopreservation technology, we will probe into the cryopreservation of cord blood and umblical cord derived stem cells, and the improvement on the process and quality management for cryostorage of stem cells and its related resources.On the cryopreservation of cord blood resources, it is a fact that the cryopreserved cord blood has been the important resources for the clinical HSC transplantation. The application of cord blood is pushing the development of the cord blood bank. Till the September of 2014, there are 167 autologous cord blood banks with 1.3 million cord blood units,45 public cord blood banks with 450 thousand cord blood units. And the ratio of cord blood transplantation in hematopoietic stem cell transplantation has been more than thirty percent. Since the beginning of cryostore umbilical cord blood, each of the preparation technology of improvement or the application of new equipment has simplified the umbilical cord blood preparation and improved the efficiency and quality of it. Through analysing and studying the application of the independently invented kashi clamp in the current work of Shandong province cord blood bank, we evaluated the application of new technology for preparation of cord blood. Besids, in order to validate the deep cryopreservation technology of cord blood for a long time, we did experiments on the influence of low temperature preservation time on the cord blood stem cell components. The experimental results showed that there was no significant difference on the number of MNC, HSC (CD34+cells) and its recovery of immune cell percentage changes, accourding to cryostorage of cord blood stem cells for 1 month and 6 months. There was the losses of nucleated cells because of the cryopreservation, but there was no significant difference on the rate of the living cord blood cells, whether storage for 1 month or 6 months.The umbilical cord derived MSC (UC-MSC) is convenient to collective, relatively pure, richfull and low immunogenicity, and therefor it becomes a important goal of cryopreservation for stem cells. At present we cryostored the MSC from the umbilical cord in order to to preservate the umbilical cord for long time, with a long time preperationg, a high cost and a poor production efficiency. If we develop a preparation solution on cryopreservation of the umbilical cord tissue, we can store a umbilical cord with many kinds of stem cells within 2-3h, preperating the MSC from it if necessary. So we explored to set up a solution for the cryopreservation of the Wharton’s Jelly from the unblical cord, and we got a preliminary optimizing protective system of cooling and freezing method.The cryopreservation of stem cell resources is a ever green engineering, It is imperative to improve the control of the process quality for the stem cells cryostorage. In the past, we managed the storage of stem cell using a traditional biological laboratory management way. Then there was no quality management system, and quality tracking system was not perfect. Now, in accordance with the GMP and ISO9001 management principles, we set up a quality management system for the quality of optimization of collecting, transport, preparation and storage the stem cell, with a quality control and technical record system, which can ensure the traceability of production preparation process and the reliability of the product quality.Understanding the present status of the deep cryopreservation technology, we find that the deep cryopreservation technology is important for storage of the cord blood stem cells, umbilical cord tissue with many stem cells in. But we should know that there are many places to be perfect and development on the application of the deep cryopreservation technology, especially in the preparation of stem cells, stem cells related to tissue preservation program and quality management. Here we hope to do some exploratory research on the above to support and safeguard the cord blood and umbilical cord resources storage.
