Objective Through establishing the rats model of osteoarthritis, under the d-fuzi intervention, observation the behavior effects of d-fuzi in the rat of osteoarthritis, discuss the possible mechanisms in the treatment of osteoarthritis of the d-fuzi.Methods The forty-eight SD rats, aged eight weeks, were randomly divided into three groups:blank group, model group, d-fuzi group. and sixteen rats in each group. No treatments to the blank groups, and others through were injected with iodine acetic acid (1.25g) in articular cavity to establish osteoarthritis animal model. Preparation of d-fuzi according to previous research method. The d-fuzi group were given the d-fuzi recipe through intragastric administration, Blank control group and model group were given physiological saline a through intragastric administration. after four weeks, Holding power test, spontaneous activity, pain response and fatigue test; After that, all rats were executed to conduct by Radiographic analysis and histopathologic analysisto take on both sides of rats knees.Result (1)histopathologic analysis:In model group, the tibial cartilage were severely damaged, therelack of cartilage, osteophyte formation, subchondral bone exposure, and sclerosis of subchondral bone, trabecular bone area to reduce. Compared with model group, In d-fuzi group, the damage articular cartilage were reduced, therelack of cartilage were not seen obvious bone, part of reduce the trabecular bone, sclerosis of subchondral bone were not clearly seen. (2)radiographic analysis:In the model group, articular cartilage degeneration was obviously, articular were taken on the bone dissolution, bone hyperplasia, cartilage surface uneven.Compared with model group, In d-fuzi group, the articular cartilage degeneration was not obvious, articular cartilage surface is smooth, the joint space is normal.(3)hold power test analysis: holding power value of the d-fuzi group and normal group was significantly higher than model group(P<0.01).(4)spontaneous activity analysis:compared with the normal group, the model group was significantly reduced in number of spontaneous activity(P<0.01); in the d-fuzi group, function of spontaneous activities was significantly recover(P<0.01.compared to model group), but it is not completely reached normal levels.(5) fatigue test analysis:the swimming time of rats of normal group and model group was basically same, but the swimming time of d-fuzi group was longer than the model group and normal group(P<0.05), du-fuzi can strengthen activity ability of the rats because of its warm drug properties.(6) pain response analysis:Pain tolerance of model group rats was decreased obviously than normal group and d-fuzi group(P<0.01), pain threshold level of the d-fuzi group almost reached to normal range or higher than the normal group after treatment(P<0.01,compared to model group; P>0.05, compared to normal group).Conclusion D-fuzi has a certain protective effect to cartilage of osteoarthritis rats, can significantly improve behavior ability of osteoarthritis rats, and has a certain treatment effect to osteoarthritis.
