Objective:To investigate the association between cervical cancer and 3 genetic single nucleotide polymorphisms(SNPs):FGFR3 C249G, XRCC1 G399A,TP53 C72G.Methods:We screened genes by protein interaction network and determined by NCBI and AmiGO and Biogrid.The participants enrolled included 102 cervical cancer patients and 163 healthy controls from The First Affiliated Hospital of Chongqing Medical University between May 2013 and January 2015.SNPs were genotyped by MAMA-PCR and direct sequencing.Results:Logistic regression analysis revealed that women carrying Gln399Gln(AA) genotype had a significantly 2.055-fold increased risk for cervical cancer [95%CI=1.039-4.016;P=0.038].Arg/Gln(GA) or Gln/Gln(AA)genotype were also had an increased risk for cervical cancer[OR=1.829;95%CI=1.063-3.148;P=0.029],compared with women carrying Arg/Arg(GG) genotype.The AA genotype had a 7.419-fold[95%CI=2.079-26.477,P=0.002] increased risk for cervical cancer, (referent, old controls).There was an increased OR[2.663;95%CI=1.316-5.390;P=0.006] for GA+GG genotype in cervical cancer cases compared to old controls.The frequency of the XRCC1 A allele in cases was significantly higher than that in old controls[OR=2.780;95%CI=1.478-5.227;P=0.001].Analysis of FGFR3 C249G and TP53 C72G polymorphisms showed no significant differences between cervical cancer patients and controls(P>0.05).The frequency of the GCGA genotypes in cases was sigficantly higher than that in old controls[OR=9.143;95%CI=1.899-44.011;P=0.006].In addition,the GGAA genotype was associated with an increased risk for the development of cervical cancer[OR=12.571;95%CI=1.280-123.480;P=0.030].Conclusions:We showed an association between the genotype of Gln/Gln (AA) or Gln/Arg (GA) and an increased risk for cervical cancer in a polulation of Chinese women.Moreover, TP53(C72G)and FGFR3(C249G) polymorphisms may not be a genetic risk for cervical cancer in our study polulation.These data suggest that there are significant associations between the G72G site SNP of TP53 and the XRCC1(A399A) polymorphism with cervical cancer in Chinese,and there is a gene-gene interaction.
