| Objective:Through the eatablishment of rabbit ears simple,to investigate the effect of 5-fluorouracil on hypertrophic scar in rabbit ears.Methods:The hypertrophic scar models in rabbit ears were reproduced and randomly divided into three groups (control group, local injection of 5-FU group and 5-FU ointment group). On postoperative 60th days, Hematoxylin-eosin and MASSON staining staining was applied to observe themorphological and collagen change of scars under light microscope. RT-PCR and ELISA were conducted to detect the expression of TGF-βã€PDGFã€bFGFã€and INF-γ.Results:HE and Masson staining showed that thinner dermis, fewer fibroblasts and collagen fibers, collagen fibers arranged regularly and less collagen nodule was observed in local injection of 5-FU groupcompared with the control group and 5-FU ointment group. Furthermore, we found that TGF-β, PDGF, bFGF, INF-γ mRNA and protein expression levels were significantly lower in local injection of 5-FU group and 5-FU ointment group than the control group, and the local injection group was more effective than the ointment group.Conclusion:5-fluorouracil is effective for the treatment of hypertrophic scars,and the localized injection of 5-fluorouracil is more effective than the ointment group. |
