| ObjectiveUse human serous papillary Cystadenocarcinoma cell SK-OV-3 to establish the model of nude mice ovary cancer transplanted tumor. Through observing the tumor-bearing mice and the growth of tumor to analyze some factors about the death of the tumor and to investigate the effect of Compound Da Qiqi Decoction combined with DDP on the growth and the relative apoptosis factors in the nude mice ovary cancer subcutaneous transplanted tumor, providing theory basis for clinical treatment.MethodFoster the SK-OV-3 cell and Inoculation it on the BALB/C nude mice to establish the model of ovary cancer transplanted tumor. Then divide the 40 tumor-bearing mice into the control group(saline group), the low dose CDQD group(12g · kg-1),high dose CDQD group(24 g · kg-1),DDP group(0.003 g · kg-1 and CDQD combined with DDP group(12 g · kg-1,0.003 g · kg-1) at random. Next step,give each group respectively treatment to observe the general condition,measure the tumor’s volume and weight,draw the curve graph of the tumor’s growth,calculate the inhibition rate of tumor.Use HE staining to observe the tumor tissue,and use Immunohistochemistry SP method to test the protein expression of Survivin,Caspase-3 and use RT-PCR along with Western Blot to test the expression of Bcl-2, Bax,Survivin andCaspase-3 mRNA and protein.Result1.The rate of establishing the model of nude mice ovarian cancer transplanted tumor reaches 100%.2.During the treatment,the condition of the tumor-bearing mice in the group with CDQD is better,indicating that CDQD combined with DDP can inhibit the proliferation of tumor obviously and the inhibition rate of tumor compared with other group has significant difference on statistics (P<0.01);and compared with the control group,the curve of tumor’s growth in each group has significant difference (P<0.01).3.HE staining shows that the tumor organization cell in negative control group has large amount and the core is big and has obvious heteromorphism and mitosis.Compared with the control group, the number of cells in other groups decreased more or less,and the tumor has less mitosis and some of them begin to shrink, showing signs of vacuolar degeneration.The result of Immunohistochemistry SP shows that the Survivin protein expression in other groups deceases clearly(P<0.01) and the Caspase-3 protein expression increases greatly(P<0.01) compared with the control group.4.The result of RT-PCR and Western Blot suggests that Bcl-2,Survivin mRNA and protein expressions decrease in other groups compared with the control group. However,the Bax,Caspase-3 mRNA and protein expressions increase and the difference is of great meaning to statistics(P<0.01).The Bcl-2,Survivin mRNA and protein expression is the lowest in CDQD combined with DDP group and the the difference was statistically significant (P<0.01).Conclusion1.CDQD has great influence on improving the general condition of the tumor-bearing mice,which is likely to be connected with the traditional chinese medicine with reducing the poison.2.CDQD has inhibiting function to ovarian cancer subcutaneous transplanted tumor’s growth. But the effect will be influenced if it is used alone without DDP,which shows that it is linked with accelerating the apoptosis of tumor cells and enhancing the sensitivity of DDP. |
