Objective: In our country, cancer is the second disease mortality and its etiology has not been fully elucidated. Hepatocellular carcinoma(HCC) is one of the most common malignant tumor, serious threat to the human health; At present, due to the lack of specificity in the diagnosis of early indicators, nearly 80% of the patients are diagnosed as liver cancer is belong to the middle and late period and lose the chance of radical excision, postoperative recurrence and transfer rate is high, more than 5 years survival rate is only 10%. Early discovery, early diagnosis and early treatment is still the top priority. The early diagnosis of liver cancer is still the best way to improve the long-term survival rate. Now,some scholars believe that cancer is also a kind of autoimmune disease. This project intends to analyze the correlation of some kinds of tumor related antigen level of autoantibodies and the development of the tumor, looking for early diagnosis of tumor markers.Method: In this study,1 immunomodulatory factors interleukin-2 receptor subunit alpha chain and C-myc cancer associated gene were selected,The bioinformatics database and software were used to analyse their structures and characters. Design and synthesis of peptide sequence as antigen, using the optimization of the double antibody sandwich ELISA method;Detection of 80 liver cancer patients and 104 healthy controls individual antitumor associated antigen autoantibodies Ig G levels in peripheral blood using SPSS20.0 software for X2 test and ROC curve analysis.Result: Overall data comparison: two groups of samples autoantibodies horizontal comparison, CD25 autoantibodies, examined the Mann-Whitney U, Z = 3.215, P = 0.001, prompt CD25 autoantibody level were difference in two groups.C-myc autoantibodies, examined the Mann- Whitney U, Z = 0.807, P = 0.420, prompt C-myc autoantibody levels had no significant difference in two groups。Compare two groups of level of autoantibodies in men sample CD25 autoantibodies, examined the Mann- Whitney U, Z = 3.024, P = 0.002, prompt CD25 autoantibody level were difference in two groups.C-myc autoantibodies, examined the Mann- Whitney U, Z = 0.487 P = 0.626, prompt C-myc autoantibody levels had no significant difference in two groups of males.Compare two groups of level of autoantibodies in women sample;CD25 autoantibodies, examined the Mann- Whitney U, Z=-1.549,P=0.121, prompt CD25 autoantibody levels had no significant difference in two groups of males.C-myc autoantibodies, examined the Mann- Whitney U, Z=-1.099,P=0.272, prompt C-myc autoantibody levels had no significant difference in two groups of males.Conclusion: CD25 Ig G autoantibodies in patients with hepatocellular carcinoma(HCC) expression in plasma is significantly higher than the healthy controls,,especially for the male patients; So the CD25 Ig G autoantibodies may has important value in the early diagnosis of hepatocellular carcinoma. C-myc Ig G autoantibodies in patients with hepatocellular carcinoma(HCC) expressions in plasma is the same as the healthy controls,so it cannot be an index of early diagnosis of hepatocellular carcinoma(HCC).
