Antrodia cinnamomea is a species strain of Taiwan and usually growing in more than 100 years of decay stem of Cinnamomum kanehirai Hay. Studies show that Antrodia cinnamomea has a wide range of biological activities, including anti-cancer, anti-inflammatory, antioxidant, antiviral, anti-fatigue, protecting liver and immune regulating activity, especially in liver disease has significant effect. Due to the demand of wild Cinnamomum kanehirai Hay is increasing. the severity of deforestation and the slowly grows of Antrodia cinnamomea are greatly increased the difficulty obtaining wild Antrodia cinnamomea. The artificial culture of Antrodia camphorata provides solutions for this dilemma. The paper consists of three parts, the first part optimization of cultivation medium ingredients of Antrodia cinnamomea to upgrade of Antrodia cinnamomea fermentation level. Subsequently, to study on the mechanisms of Antrodia cinnamomea in anti-fatigue and the protective effect on acute alcoholic liver injury.Firstly, the optimal concentration of four significant factors was optimized by using chemometric Methods and response surface analysis in this paper. An optimal formula of medium for shake culture was got(g/L), glucose 20.00, peptone 14.30, yeast extract 10.00, KH2PO4 0.74, VB1 0.17, Magnesium Sulfate 1.46 anhydrous. In a word, a mean value of desirability values Dv = 0.389 was obtained, which was 19.54% higher than the value achieved by the basal medium. The biomass, the adenosine, the content of polysaccharide and the triterpenoid were enhanced by 2.20%, 4.17%, 17.17% and 19.12%compared with the without optimization, respectively.Then through the spontaneous activity experiments, it is possible to learn that Antrodia cinnamomea may not have inhibitory or excitatory effects on the central nervous system. Through mice endurance running test, endurance rotarod test and weight loading swimming test, it has been regarded as an important indicator to reflect the endurance. In this chapter, we can know that Antrodia cinnamomea can significantly improve the physical strength and endurance of both female and male mice, and has the effect of anti-fatigue. High-intensity exercises will decreases the storage of energy materials, such as liver glycogen, muscle glycogen and ATP. The results show that Antrodia cinnamomea can prevent energy substance levels loss, before and after swimming. At the same time, Antrodia cinnamomea can inhibit the activity of lactate dehydrogenase and alleviate the accumulation of lactic acid, before or after swimming. A large number of studies show that load exercise for a long time, the body free radicals and lipid peroxidation were increased, Antrodia cinnamomea can not only reduce the MDA level, before and after swimming, but also can activity the antioxidant enzymes SOD and GSH-Px, and scavenging free radicals to alleviate the oxygen metabolism ability reduce. By Western blot analysis can be drawn, Antrodia cinnamomea to enhance Akt phosphorylation, furthermore up-regulates of AMPK phosphorylation, thereby inhibiting the expression of p-m TOR expression and alleviate the energy consumption.Large quantities of alcohol over a short time consumes, will cause the body’s alcohol overload, resulting in lipid peroxidation and inflammatory reaction and result in liver fibrosis and liver cirrhosis. Firstly, through expose alcohol over a short time to establish the model of acute alcoholic liver injury in mice. The results shown that Antrodia cinnamomea can significantly ease ALT and AST levels increased caused by alcohol induced and alleviate liver injury. Secondly, Antrodia cinnamomea can inhibit the level of NO and ROS increased induced by alcohol. In this study, the activity of SOD and GSH-Px were selected as the measuring factors. While it is not difficult to see that, Antrodia cinnamomea can increase the activity of antioxidase. Therefore, Antrodia cinnamomea can not only reduce the levels of oxidation factor, also can activate the antioxidant system, to restrain the alcohol induced lipid peroxidation. Finally, the results in this paper also confirm the role of Antrodia cinnamomea, which not only can effectively alleviate the elevated levels of TNF- alpha induced by alcohol. At the same time, Antrodia cinnamomea can also ease anti-inflammatory cytokine IL-10 levels, reduce inflammation, exert the hepatoprotective effect.
