Objective:To detect the human papillomavirus (HPV) DNA, interleukin 17A(IL-17A) gene G-197A, interleukin 17F(IL-17F) gene A-7488G and monocyte chemoattractant protein-1 (MCP-1) gene A-2518G single nucleotide polymorphism (SNPs) of epithelial ovarian cancer (EOC) tissue, and to explore the possible association and clinical significance between the HPV infection, MCP-1 and IL-17 with ovarian cancer development in Chinese Han population of Hunan region.Methods:This study was conducted with a case-control design including 130 patients confirmed by clinical diagnosis. There were 92 ovarian tissues of EOC cases being selected as case group, and 38 normal ovarian tissues cases as control group. Polymerase chain reaction (PCR) was used to detect the HPV DNA in all tissue samples, and then organizations the HPV positive rate between the two groups. The expression of the IL-17 A gene G-197A, IL-17F gene A-2518G and MCP-1 gene A-7488G single nucleotide polymorphisms of the EOC was analyzed with Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).Results:1、The positive rate of HPV DNA in the case and control group were 63% (58/92) and 23.7% (9/38), respectively. There was significant difference between the two groups (χ2=16.68, P<0.001, OR=5.5).2、IL-17A gene G-197A single nucleotide polymorphisms all have GG, AG; AA genotypes in case and control group. Compared with the control group, the frequencies of GG (13.05% vs 5.27%), AG (65.21% vs 63.15%) and AA (10.87% vs 5.26%) genotype were not significantly different in case group (χ2=2.587, P=0.274). IL-17F gene A-7488G single nucleotide polymorphisms all have AA, GA, GG genotypes in case and control group. Compared with the control group, the frequencies of AA(10.87% vs 5.26%), GA(75.00% vs 89.48%) and GG(14.13% vs 5.26%) genotype were not significantly different in case group(χ2=3.459, P=0.177). Multivariate logistic regression analysis indicated that there was no significant correlation between IL-17A gene G-197A, IL-17F gene A-7488G polymorphism and EOC(P> 0.05).3、MCP-1 gene A-2518G single nucleotide polymorphisms all have AA, AQ GG genotypes in case and control group. Compared with the control group, the frequencies of AA(17.40% vs 15.79%), AG(44.56% vs 52.63%) and GG(38.04% vs 31.58%) genotype were not significantly different in case group(χ2=0.725,P=0.696). Multivariate logistic regression analysis indicated that there was no significant correlation between MCP-1 gene A-2518G polymorphism and EOC(P>0.05).Conclusion:1、Our datas suggested that HPV infection related to the occurrence of EOC, and played a role in ovarian tumor development process in the Chinese Han population of Hunan region.2、Our datas suggested that IL-17A gene G-197 A, IL-17F gene A-7488G and MCP-1 gene A-2518G polymorphism not related to susceptibility to EOC in the Chinese Han population of Hunan region.
