Objective:Gout is a disease redused by purine metabolic disorder and UA discharge reducing.It lead to blood UA increased. In recent years,the incidence of gout is on the rise.Rational utilization of the rich resources of traditional Chinese medicine (TCM) is the new trend for the treatment of this disease.This experiment research by weighing the weight of HUA model mouse, determine the concentration of serum UA, SCr, BUN and observing related viscera under light microscopy, preliminary discussion the effection of polygonum orientale compound prescription on serum and related viscera of HUA model mouse.Method:Male,SPF,ICR mouse, fed in the laboratory freely for a week, to adapt to the environment, divide into 5 groups began the experiment, groups are blank group, model group, allopurinol group, low dose group, high dose group. Between the start of the experiment to the end of the experiment,weighing the weight of the mouse. The water extract of the polygonum orientale compound prescription crushed into powder, then dissolve into saline, make into low dose group, high dose group. Allopurinol crushed into powder, then dissolve into saline,make into allopurinol group.The period of intragastric administration is 14 days.In order to maintain a stable serum drug concentration, at the 14 day, adapt the method of intraperitoneal injection to make hypoxanthine model mouse,injection dose was 0.01 mL/g.Collect blood, after set the blood for a while, extract serum to determine the serum UA,SCr,BUN.The viscera morphological sturcture observed under the light microscope.To investigate the effect of polygonum orientale compound prescription on serum and related viscera of HUA model mouse.Results:Using the statistical software, analyze the concentration of weight,Serum UA, SCr and BUN.The viscera morphological sturcture observed under the light microscope.1、Weight changes:the blank group present normal growth; Model group and allopurinol group have growth but slow;Low dose group and high dose group have no obvious growth.2、Serum changes:compared serum UA concentration, model group significantly higher than blank group,there were significant differences; allopurinol group, low dose group, high dose group are inferior to model group; Compared SCr, BUN, there were significant differences between low dose group, high dose group and blank group.compared with model group, were higher than model group, the difference is obvious.Allopurinol group have no difference with blank group.3、Observe viscera morphological sturcture under the light microscope:heart, liver, spleen, lung were no obvious abnormalities; kidney: model group, low dose group, high dose group had inflammatory reaction, inflammatory cell infiltration around glomerular. Blank group and Allopurinol group basically showed normal renal structure.Conclusion:Intraperitoneal injection of hypoxanthine can successfully copy HUA mouse. Hyperuricemia model, Fructus polygoni orientalis compound prescription inhibite the weight of HUA model mouse and has certain damage to the kidney.
