Objective:Through the observation of different frequency electroacupun-cture stimulation in rats with cerebral infarction of the " LI 10" and "SJ 5 "on the expression of rat forelimb motor function and ischemic lesions GAP-43 and synaptophysin (SYP), and to evaluate the upper extremity motor function of cerebral infarction rat’s behavior, to further explore the effect of Electroacupuncture at different frequencies on cerebral infarction, treatment optimization scheme.Methods:The middle cerebral artery (MCAO) occlusion cerebral ischemia model was made by thread embolism method. SD male rats were randomly divided into blank group, sham operation group, model group,2Hz group,50Hz group, 100 groups of Hz, each group of 10. The model 2 weeks ago to take the pre training to grab the front of the fore. The neurological function of each rat was scored by Longa after the model 2h, and the score was 1-3. Model group, sham operation group and blank group were treated with electroacupuncture. Electroacupuncture group using 2Hz,50Hz, 100Hz and continuous wave current for the treatment of 1mA. Each group according to the postoperative 1d,3d,7d,14d,21d five time points, by screen test for evaluation of nerve function, for each time period between groups and within groups were observed and compared. All groups were sacrificed at 21 d after the experiment, and the expression of SYP and GAP 43 in the cerebral cortex were observed by immunohistochemistry.Results:1 Comparison of behavior:the results showed that:the blank group, rats in the sham operation group were screen test score of 0 and prompt operation does not affect the neural function of rat; data in each group and blank group compared with statistical significance (P<0.05). On postoperative day 1 and 3, compared with model group, EA group,50Hz and 100Hz groups, the difference was not statistically significant (P>0.05), model group, EA group,50Hz and 100Hz group were compared with control group, the differences have statistical significance (P<0.01). On the 7 day after surgery, there was significant difference between the 50Hz group and the model group (P<0.01). After 14 days,50Hz group and model group compared with statistical significance (P<0.05); 50Hz and 100Hz group compared with statistical significance (P<0.05); group 2 Hz and 50 Hz compared with statistical significance (P<0.05).21 days after operation,2Hz group and model group compared with statistical significance (P<0.05); 50 Hz group and model group compared with statistical significance (P<0.01); group 2 Hz and 50 Hz compared with statistical significance (P<0.01); 50Hz and 100Hz group compared with statistical significance (P<0.01).2 SYP:the expression of SYP decreased significantly after cerebral infarction, the expression of SYP was significantly increased after electroacupuncture treatment. Sham operation group,50Hz group were compared with the blank group (P>0.05); the rest of the group and the blank group comparison, (P< 0.01); 2Hz group and model group comparison, (P<0.05).50Hz, 100Hz group were compared with the model group, (P< 0.01); 2 Hz and 50 Hz group comparison, P< 0.01; 100Hz group and EA group, (P< 0.05). GAP-43:the stress of GAP-43 after cerebral infarction was increased, and then decreased gradually, and electroacupuncture treatment could keep the GAP-43 at a high level. Sham operation group and blank control group, (P>0.05), the rest of the group and the control group compared with statistical significance (P<0.01); 50Hz group and model group comparison, (P<0.01); 2Hz group,100Hz group were compared with the model group (P>0.05); 50Hz and 100Hz groups were compared and 2Hz group, (P<0.05),50Hz and 100Hz group comparison, (P< 0.01).Conclusion:1.Different electric acupuncture frequency stimulation can promote the recovery of neurological function of cerebral infarction,50HZ group is better than 2Hz group, 100Hz group of electroacupuncture group.2.Different electro acupuncture frequency stimulation can promote the expression of SYP and GAP-43 protein in cerebral cortex after cerebral ischemia, and the 50Hz group was obviously.3.Tthe promoting effect of electroacupuncture at different frequencies on the recovery of neurological function after cerebral ischemia may be related to the increase of SYP and GAP-43 protein expression.
