Objective:On the basis of preliminary research,To study the impact on HPOA morphology and ultrastructurethe in Ovulatory disorder rat caused by Kidney Deficiency by Tongmaidasheng-Tablet. And combine with changes in hormones, to provide a theoretical basis and scientific evidence for BushenTiaoChong.Method:According to the method of Yujin.ASR model was established by injecting testosterone propionate subcutaneously to SD female rats on 9 days age Then the rats were divided into seven groups:model group,TMDST high-dose group,TMDST medium-dose group,TMDST low dose group,letrozole group,You gui-pill group and blank control group.The medication was started at 80 days old until 5 weeks later,then the blood was taken through femoral artery.The level of serum FSH、LH、E2 were detected by Radioimmunoassay, then the Brain、pituitary、bilateral ovaries and Uterus were stripped quickly for inspection.Results:1.HypothalamicUnder the light microscopy:The hypothalamic arcuate nucleus neurons area and the content of nissl bodies of model group are smaller than blank group (P<0.01); Compared with model group,the high and middle-dose group, letrozole group are larger obviously(P<0.01),the low-dose group、yougui-pill group also larger (P<0.05).2. PituitaryUnder the light microscopy:the adenohypophysis structure is sparse,and the pituitary nerve cells area of model group are smaller than blank group (P<0.01); the highu middle, low-dose group, letrozole group are larger obviously compared with model group (P<0.01),the yougui-pill group also larger (P<0.05).Under the electron microscope:In model group,the form of Gonadotroph cell is irregular,in cells nucleus can be visioned, nucleus gap widened, Endoplasmic reticulum expansion and show cavitation model change. Ribosome is missing in the cell. Mitochondria swelling and less secretory granules.Compared with model group,structureall of all the treatment group have different levels improvement.3. OvaryUnder the light microscopy:①in model group, the number of all levels follicles are less than blank group (P<0.01);compared with the model group,The high, middle-dose group、letrozole group are Significantly increased (P<0.01).The yougui-pill group are also increased (P<0.05).②in model group,the adenohypophysis structure is sparse,and the Granular cell layer is thinner, the area of ovarian granulosa cells are smaller than blank group (P<0.01); compared with the model group,all treatment group are lager obviously (P<0.01).③in model group,the follicles showed a form of pathologic cystic expansion, the number are more than blank group (P<0.01), compared with model group,the high、middle-dose group、yougui-pill group、letrozole group are less.Under the electron microscope:in model group,the shape the ovarian granulosa cells is irregular,the nucleu is crumb,the ribosome is lost,mitochondria is swelling and can be seen apoptotic bodies; compared with model group,each treatment group Granular cell ultrastructure all have different degrees of improvement.4. UterusUnder the light microscopy:The uterine glands ares and the endometrium thickness of model group are smaller and thinner than blank group (P<0.01); compared with model group endometrium,the high-dose group are obviously lager (P<0.01),the middle,low-dose group and letrozole group alsoUnder the electron microscope:The endometrial epithelial cells are minute-level distribution, the shape of nuclear is Irregular.Compared with model group,each treatment group Granular cell ultrastructure all have different degrees of improvement.5. HormoneIn model group,the level of FSH、LH、E2 were lower than those in normal group (P<0.01); Compared with the model group,the level of FSH is higher in high-dose group (P<0.01),the midd、LH-dose group are also higer (P<0.05).The level of LH is higher in all treatment group (P<0.01).The level of E2 are higher in high、middle-dose group (P<0.01)Conclusion:1.Given intramuscular injection of testosterone propionate method successfully reproduce the type kidney ovulation disorder infertility in rats (ASR) model. The model can lead to a mature follicle and corpus luteum formation and pathologic cystic follicles.2.TMDSP can influence HPOA of model rats, morphological structure and cell ultrastructure to adjust its endocrine function, thus promote the follicular development and ovulation.3.TMDSP acting on the endometrium, can restore the endometrial epithelial cell ultrastructure, promote the endometrial thickening, the secretory transformation, thus increasing the receptivity of endometrium. Is conducive T implantation.
