Solanum nigrum L., a kind of perennial herb, also known as bitter herbs, bitter kwai or tulipa eyed grass, is one of the Solanaceae species that are distributed throughout the whole country. The Compendium of Materia Medica recorded that Solanum nigrum L. is bitter, cold and non-toxic with detoxification and detumescence. At present, little research has been done on the hepatoprotective effects of Solanum nigrum L.To provide scientific basis for the clinical application, the hepatoprotective activity of Solanum nigrum L. has been systematically studied. There are 3 parts of experimental research as follows:Part 1:The water-soluble polysaccharide, the alkali-soluble polysaccharide, the total organic acid, the total alkaloid and the total saponin are extracted from Solanum nigrum L. And the content of these extractions is determined by the UV spectrophotometry method. The result is (83.26±3.51)%, (69.04±2.88)%, (59.36±1.40)%, (69.25±2.11)% and (65.57±4.52)%.Part 2:An animal model of acute liver injury in mice induced by CCl4 is established to evaluate the hepatoprotective activities of these fractions. Through the detection of ALT, AST and ALP in the serum, the water-soluble polysaccharide and the total saponins are proved to be the pharmacological active fractions.Part 3:In our systematical study on the chemical constituents of solanum nigru m L.,12 compounds were obtained. Their structures were elucidated by the basis of physic-chemical property and spectroscopic analysis. The 12 compounds are β-sitost erol, β-daucosterol, qercetin, quercitrin, isoqueritrin, quercetin-3-O-β-D-glucop-yr anosyl(1→2)-p-D-glucopyranosid,8-O-β-D-glucopyranosyl betulinic acid 3β-O-βD-gl ucopyra-noside, β1-solasonine, β2-solasonine, (3β,22a,25R)-spirosol-5-en-3-yl-O-α-L-r hamanop-yranosyl-(1→2)-[O-β-D-glucopyranosyl(1→3)]-O-P-D-galactopyranoside, (3β,2 2a,25R)-spirosol-5-en-3-yl-O-a-L-rhamanop yranosyl-(1→4)-OβP-D-galactopyranoside, (3β,22a,25R)-3,12-dihydroxy-spirosol-5-en-oic acid。... |
