Objective:This study aims to detect the expression of S100A8 and ASAH1 in non-invasive breast ductal carcinoma before neoadjuvant chemotherapy, and to investigate the correlation between their expression and chemotherapy response, or other clinicopathological parameters.Methods:Miller & Payne stage was used to assess the chemotherapy response of breast cancer tissues. Patients were divided into a chemoresistant group (grades 1-2) and a chemosensitive group (grades 3-5) according to M & P grades. The detection of ASAH1 was used Elivisionplus method and the expression of S100A8 was examined using Roche closed immunohistochemical staining system. Positive and negative controls were also used in this study.Results:1) Total 120 cases.60 cases from Qilu Hospital (group â… ), the other 60 cases from Liao Cheng People’s Hospital (group â…¡). there were 57 chemosensitive (57/120, 47.5%) and 63 chemoresistant (63/120,52.5%). Of that,20 cases was chemosensitive (20/60,33.3%) and 40 cases was chemoresistant (40/60,66.7%) in group I. In group â…¡,37 cases was chemosensitive (37/60,61.7%) and 23 cases was chemoresistant (23/60.38.3%).2) In group â… , S100A8 positive expression was showed in 21 cases (21/60,35%), including 12 chemosensitive (12/20,60%) and 9 chemoresistant (9/40,22.5%). In group â…¡, S100A8 positive expression was showed in 26 cases (26/60,43.3%), including 19 chemosensitive (19/37,51.4%) and 7 chemoresistant (7/23,30.4%). Total 120 samples, the positive expression of S100A8 was observed in 47cases (47/120,39.2%), consisting of 31 chemosensitive (31/57,54.4%) and 16 chemoresistant (16/63,25.4%). There was a significant difference of S100A8 expression between chemosensitive and chemoresistant groups (P< 0.05).3) The positive expression of ASAH1 was observed in 18 cases (18/60,30%) in group â… , including 5 chemosensitive (5/20,22.5%) and 13 chemoresistant (13/40,32.5%). In group â…¡,19 cases (19/60,31.7%) were showed the positive expression of ASAH1, consisting of 12 chemosensitive (12/37,32.4%) and 7 chemoresistant (7/23,30.4%). Total 120 samples, the positive expression of ASAH1 was observed in 37cases (37/120,30.8%), consisting of 17 chemosensitive (17/57,29.8%) and 20 chemoresistant (20/63,31.7%). There was no significant difference of ASAH1 expression between chemosensitive and chemoresistant groups (P> 0.05).4) S100A8 positive expression was significantly associated with molecular subtype and histological grade. No significant correlation was found between its expression and age, tumor size, or lymph node metastasis.5) ASAH1 positive expression was significantly associated with lymph node metastasis. There was no significant correlation between its expression and age. tumor size, molecular subtype or histological grade.Conclusions:The positive expression of S100A8 significantly associated with chemosensitivity. indicating its role in breast cancer chemotherapy. S100A8 might be used as a biomarker to predict chemotherapy response. A correlation was found between ASAH1 positive expression and lymph node metastasis. ASAH1 might have important role in predicting prognosis of patients with breast cancer. Both S100A8 and ASAH1 showed their clinical value in breast cancer treatment. |
